agrochemical analysis of soil, the effect of microbial preparations onbiological characteristics and organic matter in soil was assessed aswell.There were used usual laboratory methods for the analyses.Following analyses were carried out for soil samples:- Absolute soil moisture by gravimetric method,- Content of NH 4 -N and NO 3 -N by colorimetric method withSKALAR instrument,- Production of CO 2 after 1-day incubation in thermostat at thetemperature 25 o C by the method of gas chromatography (samplewas pre-incubated 1 day in thermostat at 25 o C),- Respirometric test – basal respiration of soil CO 2 (B) andrespiration of soil with adding solution of glucose CO 2 (G) andnitrogen and glucose CO 2 (NG) after 20-hours incubation inthermostat at the temperature 25 o C (samples were pre-incubated 5days in thermostat at 25 o C).- Based on measured data there were calculated ratios G: B and NG:B, which express stability of soil organic matters (Novák, 1964)- Total nitrogen mineralization (TMN) after 14-day aerobicincubation of soil samples in thermostat at the temperature 25 o C;- Nitrification intensity (NIT) after14-day aerobic incubation of soilsamples in thermostat at the temperature 25 o C;- Total nitrogen content in microbial biomass (MB_C) byrehydration method (Růžek, 1992),- Content of Cox (Tjurin), Nt (Kjeldahl), ratio HK: FK(spectrophotometrical method), P, K, Ca, Mg (Melich III) and pH (nKCl).At the end of vegetation period measurements of growthparameters were performed in plantations, including detailedassessment of the architecture of the root system. Effect ofpreparations on the survival and following adaptation process ofspruce, fir and beech was assessed as well. Differences werestatistically tested (one-factor variation analysis, T-test). We alsotested how the application of bacterial soil conditioner affects theresistance of plantations against everywhere-present fungi Armillariasp. and Heterobasidion sp.The effect of all soil conditioners was tested also under theshelter of disintegrating spruce stands with stocking 0.6-0.7(experimental plot Oščadnica IV).
