Farm Przegl Nauk, 2009,2, 61-70 Wstęp Badania nad mechanizmem toksycznego działania glikolu etylenowego (GE) były dotąd głównie skoncentrowane na utlenianiu tego związku przez dehydrogenazy: alkoholową (ADH) i aldehydową (ALDH) do aldehydu i kwasu glikolowego. Ostatnie doniesienia wskazują, że mechanizm toksycznego działania GE może polegać również na tworzeniu wolnych rodników i ich łączeniu się z makromoleku- copyright © 2009 Grupa dr. A. R. Marcinkowskiego w poznaniu Streszczenie Badania nad mechanizmem toksycznego działania glikolu etylenowego były dotąd głównie skoncentrowane na utlenianiu tego związku przez dehydrogenazę alkoholową i aldehydową. Przemiany glikolu etylenowego do formaldehydu przebiegają przy współudziale cytochromu P450. Początkowo zakładano, że bierze w nich udział jedynie izoforma CYP 2E1. W chwili obecnej istnieją przesłanki, że w przemiany te są zaangażowane inne izoenzymy. W świetle powyższych danych celem niniejszego projektu badawczego jest kompleksowa ocena przydatności 4-metylopirazolu (4-MP) w postępowaniu leczniczym w zatruciach pojedynczych glikolem etylenowym. Badania zostały przeprowadzone na dojrzałych szczurach samcach szczepu Wistar. Glikol etylenowy w dawce 3830 mg/kg m.c. (1/2 DL 50 ) był podawany per os sondą dożołądkową, natomiast 4-MP w dawce 10 mg/kg m.c. dootrzewnowo. Materiał do badań (nerki, wątroba) był pobierany od zwierząt po 8; 12; 18; 24; 36 i 48 godzin od podania ksenobiotyku. W uzyskanych mikrosomach wykonano oznaczenia aktywności układu monooksygenaz zależnych od cytochromu P450 oraz określono poziom izoform 2E1, 1A2 i 2B1/2. Uzyskane wyniki wskazują, że 4-MP indukuje poziom cytochromu P450, praktycznie nie wpływając na pozostałe składniki układu monooksygenazowego zarówno w wątrobie jak i w nerce. Co więcej 4-MP indukował wszystkie badane izoformy P450. słowa kluczowe: CYPP450, szczur, wątroba, nerka, glikol etylenowy, 4-metylopirazol Abstract Until now the researches on the mechanism of the toxic influence of the ethylene glycol were mainly concentrated on the oxidation of this compound by the alcohol and aldehyde dehydrogenases. The changes of the ethylene glycol into formaldehyde proceed with the participation of the cytochrome P450. At first it was assumed that only the CYP2E1 isoform participates in them. At present there are premises that other isoenzymes are in these changes involved. Considering the above-mentioned data, the aim of this research project is comprehensive evaluation of the usefulness 4-methylpyrazole (4-MP) during treatment in the isolated ethylene glycol poisonings. The researches were done on mature breed of Wistar male rats. Ethylene glycol in dose 3830 mg/kg body weight (1/2 DL 50 ) was given per os with a stomach probe, whereas 4-MP in dose 10 mg/kg body weight into peritoneum. The research material (kidneys, liver) was taken from animals 8; 12; 18; 24; 36 and 48 hours after giving the xenobiotic. In resulted microsomes there were done the markings of the activity of the monooxygenase system depending on the cytochrome P450 and there was also determined the level of the isoforms 2E1, 1A2 and 2B1/2. The received results indicate that 4-MP induces the level of the cytochrome P450, practically not having influence on the other ingredients of the monooxygenase system, both in the liver and in the kidney. Additionally, 4-MP induced all the researched isoforms P450. keywords: CYPP450, rat, liver, kidney, ethylene glycol, 4-methylpyrazole łami komórkowymi, a także bezpośrednim cytotoksycznym działaniu metabolitów. Tym samym stosowanie w terapii zatruć glikolem etylenowym jedynie odtrutek spowalniających przemiany GE za pośrednictwem ADH może być niewystarczające. Przemiany GE do formaldehydu przebiegają również przy współudziale cytochromu P450. Początkowo zakładano, że bierze w nich udział jedynie izoforma CY- P2E1. W chwili obecnej istnieją przesłanki, że w przemiany te są zaangażowane inne izoenzymy jak np. CYP1A2 61
