EXPRESSION OF E-CADHERIN AND CD10 IN PROSTATIC ADENOCARCINOMAS SUMMARY Prostate cancer is the most common malignancy among men, with a higher incidence over the age of 50. The small size of material in needle biopsies used for diagnosis of the prostate cancer coupled with the detectability of increasingly higher number of welldifferentiated cancers by the help of new methods make it more difficult for a pathologist to establish a diagnosis of prostate cancer. In addition to routine methods, some supportive methods such as immunohistochemistry are required. The antibodies used in our study are E-cadherin and CD10. In our study, we examined 171 prostate biopsy materials sent to our Division between 1986 and 2005. Gleason grading was performed to determine the histological grade of the prostate adenocarcinomas, and Gleason scores of tumours were determined. The cases were stained with the antibodies, CD10 and E-cadherin. The studies with CD10, on the basis of each parameter of Gleason score, demonstrated no staining in the cases with Gleason score 2-4 (low grade). Staining was seen in the 8-10.5 % of the cases with Gleason score 5-7 (intermediate grade) and in the 89.5 - 92 % of the cases with Gleason score 8-10 (high score). In the studies with CD10, CD10 staining was not seen in prostate tumours with glandular pattern. However, it was seen in the solid prostate tumours and the prostate 52
tumours showing single cell infiltration. On the basis of these findings, it was concluded that immunoreactivity of CD10 will help in determining the Gleason score of a tumour. In the immunohistochemical studies with E-cadherin, two of the 8 cases with Gleason score 2-4 (low grade) showed grade 1 staining, whereas two cases showed grade 2 staining and the remaining four showed grade 3 staining. Of the 84 cases with Gleason score 5-7 (intermediate grade), seven showed grade 0 staining; 16, grade 1 staining; 33, grade 2 staining and 28 showed grade 3 staining. Of the 79 cases with Gleason score 8-10 (high grade), nine showed grade 0 staining; 15, grade 1 staining; 27, grade 2 staining and 28 showed grade 3 staining. In the light of these findings, it was concluded that the immunoreactivity of Ecadherin can not be used in prostate adenocarcinoma for diagnostic purposes.
