UNIUNEAEUROPEANĂGUVERNUL ROMÂNIEIMINISTERUL MUNCII, FAMILIEI ŞIPROTECȚIEI SOCIALEAMPOSDRUFondul Social EuropeanPOS DRU2007-2013Instrumente Structurale2007-2013Investeşte în oameni!FONDUL SOCIAL EUROPEANProgramul Operaţional Sectorial pentru Dezvoltarea Resurselor Umane 2007 – 2013OIPOSDRUUNIVERSITATEADE VEST DINTIMIŞOARAThe aim of the PhD thesis, was on the one hand, to <strong>de</strong>epen the knowledge regarding the ecotoxicityof some xenobiotics: the parabens, compounds found in the environment since the early 1920s, and of whoseecotoxicity is revaluated in recent years, and, on the other hand, the ionic liquids, compounds placed on themarket recently, which have relatively few ecotoxicological information.The first part of the thesis, entitled Overview of some xenobiotics and their ecotoxicological actionsummarized the current knowledge on the concepts of ecotoxicology, the <strong>de</strong>finition of xenobiotics, notionsregarding some food and cosmetic additives and their biotransformation, notions regarding ionic liquids and theirbiotransformation. Another issue addressed in this chapter concerns the presentation of ultra performance liquidchromatography and electroanalitical methods, such as: cyclic voltammetry and cronoamperometry,simultaneously highlighting both the benefits of their use and also their disadvantages.The second part of the PhD thesis that stands for personal contributions is divi<strong>de</strong>d into the 2 mainchapters Materials and Methods and Results and Discussions, comprising a total of 10 studies. The second part ofthe thesis contains results with respect to the issues of <strong>de</strong>termination and quantification of some xenobiotics andmetabolites resulted from biotransformation and <strong>de</strong>veloping a strategy for ecotoxicological testing them at multiplelevels.There have been achieved the optimization of parabens separation by ultra performance liquidchromatography – photo dio<strong>de</strong> array <strong>de</strong>tection (UPLC-PDA) with a good un<strong>de</strong>rstanding of the effects of variousparameters on parabens extraction from cosmetic products (the nature of solvent, the homogenizationprocedure, the temperature extraction and extraction time), as well as the effects of different chromatographiccondition on the separation of compounds studied, such as: the choice of optimal <strong>de</strong>tection, the optimization ofmobile phase composition, the effects of UPLC column loading, all of them necessary for the <strong>de</strong>velopment ofnew separation UPLC-PDA methods.It was validated the <strong>de</strong>velopment of the UPLC method for qualitative and quantitative analysis ofparabens from cosmetic samples, in conformity with international requirements.Through the UPLC method there were analyzed extracts from cosmetics products purchased from thelocal market: shampoos, shower gels, body lotions, conditioners, body creams, sun creams, anti-cellulitecreams, feminine hygiene and cleansing products.There were <strong>de</strong>veloped some electroanalitical methods for amperometric <strong>de</strong>tection and <strong>de</strong>terminationof parabens in hydroalcoholic and aqueous solutions, as well as for propyl gallate in aqueous solutions with acommercial boron-doped diamond electro<strong>de</strong>.UPLC - PDA technique was used to <strong>de</strong>scribe the metabolic profile of parabens in the nemato<strong>de</strong>Eisenia foetida homogenates, to assess the biotransformation of parabens and to quantify biotransformationproducts.The second experimental direction of the study aimed to elaborate a new strategy for achievingecotoxicological battery with multiple levels.Limit tests were conducted on the fishes Poecillia reticulata and Xyphophorus helleri of commonparabens and some ionic liquids.There were performed studies at molecular level (enzyme) of some ionic liquids’ activities, studiesinvolving optimization of enzyme tests and ionic liquids testing, using multienzymatic ecotoxicological battery.Research conducted in the doctoral thesis ends with a series of Final conclusions drawn from theexperiments and is focused on personal contributions brought to the proposed theme.291POSDRU 21/1.5/G/38347”Parteneriat local pentru ridicarea nivelului şcolilor doctorale şi valorificarea potenţialului uman din cercetare îndomenii prioritare ale etapei posta<strong>de</strong>rare a României la UE”
