Paolo Di MascioLESÃO EM BIOMOLÉCULASO objetivo da nossa pesquisa é investigar osaspectos fundamentais da química e bioquímica <strong>de</strong>espécies reativas <strong>de</strong> oxigênio e <strong>de</strong> nitrogênio,particularmente, uma forma eletronicamente ativadado oxigênio molecular, o oxigênio singlete, implicadoem vários processos fisiológicos e patológicos. Maisrecentemente, temos investigado também omecanismo <strong>de</strong> ação <strong>de</strong> porfirinas tetrarutenadas,carotenói<strong>de</strong>s, flavonói<strong>de</strong>s extraídos <strong>de</strong> plantasbrasileiras. Enfatizamos a i<strong>de</strong>ntificação, osmecanismos <strong>de</strong> geração e lesões estruturais embiomoléculas e microestruturas celulares. Proteínas,lipídios insaturados e DNA são alvos biológicospreferenciais <strong>de</strong>stas espécies, disparando respostasbiológicas normais ou <strong>de</strong>letérias.SummaryOur attention focused on the chemical sourcesand the noxious behaviour of molecular oxygen/nitrogen-<strong>de</strong>rivedfree radicals in biological systems. Ourstudies have focused on i<strong>de</strong>ntifying the mechanismby which singlet oxygen ( 1 O 2) and other reactive oxygen/nitrogenspecies play their physiological andpathological roles. We have <strong>de</strong>voted efforts to <strong>de</strong>velopsuitable 1 O 2generators based on the thermolysisof endoperoxi<strong>de</strong>s. We have used this approach inour studies to <strong>de</strong>tect 1 O 2-induced damage in cells(DNA, lipids, proteins) and to screen biologically occurringcompounds for the quenching of 1 O 2. Recently,we synthesized the first water-soluble naphthaleneendoperoxi<strong>de</strong> isotopically labeled as a sourceof singlet oxygen-18.
