CHARACTERIZATION OF TWENTY DINUCLEOTIDE<br />

MICROSATELLITE LOCI FOR COMMON BEAN<br />

(Phaseolus vulgaris L.)<br />

ABSTRACT<br />

Common bean, (Phaseolus vulgaris L.), is an importante legume food in the world. We identified and<br />

characterized twenty microssatelitte loci in common bean. An enriched library was constructed and screened<br />

for two microsatellite repeat sequences (CT and GT). Microsatellite PCR-amplification was tested for 14<br />

accesions of the <strong>IAC</strong> Germplasm Bank, including Andean and Mesoamerican gene pools. The allele<br />

number ranged from one to three, and the polimorphism information content (PIC) between 0.14 and 0.65.<br />

These polymorphic loci can be used for genetic and QTL mapping, marker-assisted selection and germplasm<br />

characterization in common bean.<br />

1. INTRODUCTION<br />

Molecular markers have been used in many crop species. Common bean (Phaseolus vulgaris<br />

L.) is the principal leguminose to human consume, mainly in countries of South America and Africa<br />

(CIAT 1990). Brazil is the most important productor and consumer of bean in the world, but the crop<br />

<strong>do</strong>esn’t have the maximum producti<strong>vi</strong>ty (Silva 2000). The increase of the genetic improvement depends<br />

on the development of new molecular markers, like microsatellites.<br />
