Kuprienė, A. Blinstrubienė, V. JonytienėSummaryThe investigations were carried out in the Agrobiotechnology Laboratory of LithuanianUniversity of Agriculture. Aim of the research – to evaluate effect of co-cultivation of theexplant with agrobacterium, containing the different plasmids, on regeneration responseof winter rapeseed. Stem segments of Brassica napus co-cultivated with Agrobacteriumtumifaciens GV3101 strain containing the plasmids pART-BnMYBsn-MYBanti, pHellsgate-BnCBF and pHellsgate-BnICE. The results of the experiments indicated a difference amongthe tested cultivars regarding the<strong>ir</strong> organogenic response depended on genotype and vectorconstruction. Explants inoculation with agrobacterium containing different plasmids didn’thave any significant effect on adventitious shoots formation in cultivars ‘Libea’, ‘Liprima’(pART-BnMYBsn-MYBanti and pHellsgate-BnICE) and cultivar ‘Siska’ (pHellsgate-BnICE).The co-cultivation of explants with A. tumefaciens had a significant negative influence onshoot formation frequency of other tested cultivars. Kanamycin-resistant plant-regenerantsformation frequency strongly depended on genotype-plasmid interaction. Highest amountof resistant regenerants have been obtained after inoculation with A. tumefaciens containingpHellsgate-BnICE (cv. ‘Banjo H’); pHellsgate-BnCBF (cvs. ‘Elvis’, ‘Valesca’, ‘Libea’,‘Liprima’ and ‘Insider’); pART-BnMYBsn-MYBanti (cvs. ‘Silvia’ and ‘Siska’).Key words: Agrobacterium tumifaciens, Brassica napus, co-cultivation, kanamycinresistance,organogenesis.171
