vehicle. Brains from a subset of animals will be processed for determination<br />

of the volume and area of NMDA-injured cortex, using an image analyzer.<br />

The remaining animals will be transcardially perfused and processed for<br />

immunohistochemistry [Giampà et al. 2006], which will be performed on cortices<br />

to evaluate BDNF immunoreactivity. Immunolabeled tissue will be<br />

observed under a fluorescence microscope and images will be digitally<br />

acquired to evaluate possible labeling intensity differences between treatments.<br />

These experiments will show if the PDE inhibitor TP-10 exerts a direct<br />

neuroprotective role in the cortical neurodegeneration model of NMDA<br />

lesion, and if neuroprotection is comparable to the one achieved by PDE4<br />

inhibitor rolipram [DeMarch et al. 2007]. Moreover, the BDNF immunolabeling<br />

studies will show if neuroprotection is linked to the upregulation of<br />

BDNF. That will shed light on the possibility of using PDE inhibitors in neurodegenerative<br />

disorders where cortex is involved, such as AD.<br />
