non sono noti, ma lo stu<strong>di</strong>o <strong>del</strong>la letteratura permette <strong>di</strong> avanzare alcune<br />
ipotesi.<br />
Lo stress ossidativo, generato dalla riduzione intracellulare <strong>del</strong> metallo,<br />
sarebbe il principale responsabile <strong>del</strong>l’attivazione <strong>del</strong>le vie metaboliche<br />
che conducono all’apoptosi. Tuttavia la produzione <strong>di</strong> <strong>di</strong>fferenti<br />
ROS può indurre risposte cellulari <strong>di</strong>verse.<br />
Il destino <strong>del</strong>le cellule può essere influenzato dalla concentrazione<br />
<strong>del</strong>lo xenobiotico poiché basse dosi <strong>di</strong> esposizione sono associate ad una<br />
risposta cellulare <strong>di</strong> sopravvivenza mentre dosi più elevate facilitano la<br />
trasduzione <strong>del</strong> segnale apoptotico.<br />
La <strong>di</strong>versa efficacia e durata <strong>del</strong>l’inibizione sulle PTPs, che è funzione<br />
<strong>del</strong>la specie chimica <strong>del</strong> metallo a cui le cellule sono esposte, potrebbe<br />
suscitare risposte cellulari <strong>di</strong>vergenti.<br />
Infine bisogna considerare che le cellule provenienti da tessuti o da<br />
specie animali <strong>di</strong>verse sono dotate <strong>di</strong> apparati enzimatici eterogenei.<br />
Quin<strong>di</strong>, in presenza <strong>di</strong> un comune fattore stimolante come l’esposizione al<br />
vana<strong>di</strong>o, le cellule possono esibire una maggiore o minore efficacia sia<br />
nella genesi dei ROS sia nella capacità <strong>di</strong> contrastarne gli effetti citotossici.<br />
CONCLUSIONI<br />
Nonostante i numerosi stu<strong>di</strong> sul vana<strong>di</strong>o, non è stato ancora possibile<br />
definire il potenziale cancerogeno <strong>del</strong> metallo e rimane da chiarire la relazione<br />
tra esposizione <strong>ed</strong> apoptosi cellulare. Queste due tematiche sono<br />
intimamente connesse tra loro perché le cellule in cui il vana<strong>di</strong>o provoca<br />
un danno irreparabile <strong>del</strong> DNA e che pertanto possono subire una trasformazione<br />
cancerogena, vengono eliminate me<strong>di</strong>ante apoptosi. Tuttavia<br />
è stato osservato che il metallo possi<strong>ed</strong>e capacità anti-apoptotiche e può<br />
quin<strong>di</strong> consentire l’accumulo <strong>di</strong> cellule potenzialmente cancerogene.<br />
Appare quin<strong>di</strong> evidente la necessità <strong>di</strong> approfon<strong>di</strong>re la conoscenza<br />
dei meccanismi molecolari che <strong>di</strong>sciplinano la relazione tra l’apoptosi<br />
cellulare e l’esposizione al vana<strong>di</strong>o per ottenere una migliore comprensione<br />
dei sistemi <strong>di</strong>fensivi che le cellule utilizzano per affrontare una possibile<br />
minaccia tumorale <strong>ed</strong> al contempo per definire il potenziale cancerogeno<br />
<strong>del</strong> metallo.<br />

BIOLOGICAL MONITORING OF EXPOSURE TO BENZENE:<br />
ANALYSIS OF URINARY T,T-MUCONIC ACID IN WORKERS<br />
EXPOSED TO LOW OCCUPATIONAL LEVELS OF BENZENE.<br />

ABSTRACT. Trans, trans-muconic acid in the urine has been<br />
consider<strong>ed</strong> for many years a valid biomarker of occupational exposure<br />
to benzene. Our study cover<strong>ed</strong> a population of workers expos<strong>ed</strong> to low<br />
occupational levels of benzene. Biological monitoring has been<br />
perform<strong>ed</strong> on 371 males working in 4 <strong>di</strong>fferent <strong>di</strong>visions of an oil<br />
refining plant. We measur<strong>ed</strong> urinary concentrations of t,t-muconic acid<br />
at the beginning and at the end of the work shift. Since it is not allow<strong>ed</strong><br />
to smoke during work shift, the biological results would have not been<br />
significantly influenc<strong>ed</strong> by this habit. The mean urinary excretion<br />
of t,t-muconic acid, both at the beginning (76,32 µg/g creat) and<br />
at the end of the work shift (112,52 µg/g creat.) were similar to the one<br />
measur<strong>ed</strong> in non-expos<strong>ed</strong> population. These data confirms the<br />
effectiveness of preventive measures adopt<strong>ed</strong> in order to minimize the<br />
occupational exposure of workers employ<strong>ed</strong> in <strong>di</strong>fferent oil refining<br />
processes. However, it is important to point out that, in every <strong>di</strong>fferent<br />
<strong>di</strong>visions examin<strong>ed</strong>, the mean urinary concentrations of t,t-muconic acid<br />
at the end of the work shift result<strong>ed</strong> significantly higher than at the<br />
beginning of the shift. This observation could be due to the effective<br />
exposure to benzene, even at low concentration, in the workplace.<br />
In conclusion our data suggest that, even in the case of a low<br />
professional exposure to benzene, the urinary t,t-muconic acid is a very<br />
sensitive marker able to show minimal occupational exposure.<br />
INTRODUZIONE<br />
Il benzene è stato ampiamente utilizzato in ambito industriale, ma per<br />
le caratteristiche tossicologiche e per la comprovata cancerogenicità, è stato<br />
progressivamente sostituito da altri solventi aromatici o alifatici. Continua<br />
tuttavia ad essere impiegato in alcuni processi chimici <strong>di</strong> sintesi e nei laboratori<br />
<strong>di</strong> analisi. Anche la raffinazione <strong>del</strong> petrolio, la preparazione e la <strong>di</strong>stribuzione<br />
dei <strong>di</strong>stillati sono attività che possono comportare un’esposizione<br />
professionale a benzene. La sua presenza nei carburanti (benzine) in<br />
concentrazione prossima all’1% e, <strong>di</strong> conseguenza, nei gas <strong>di</strong> scarico <strong>del</strong>le<br />
autovetture con marmitte il cui processo <strong>di</strong> catalisi risulta esaurito, ha <strong>di</strong><br />
fatto contribuito a rendere il benzene un inquinante ubiquitario.<br />
L’anello benzenico è chimicamente molto stabile, ma, per una percentuale<br />
intorno al 2%, è prevista la sua apertura per formare un metabolita,<br />
l’acido trans, trans- muconico, non dotato <strong>di</strong> tossicità <strong>ed</strong> escreto con le<br />
urine. L’acido t,t-muconico urinario ha acquistato rilevanza crescente nell’ultimo<br />
decennio in quanto si è rilevato un metabolita utilizzabile come in<strong>di</strong>catore<br />
per il monitoraggio biologico <strong>del</strong>l’esposizione a benzene (1, 2).<br />
Tuttavia, nell’impiego <strong>di</strong> questo in<strong>di</strong>catore per il monitoraggio biologico<br />
<strong>del</strong>l’esposizione occupazionale a bassi livelli ambientali <strong>di</strong> benzene,<br />
risultano essere rilevanti le interferenze rappresentate dalla quota<br />
urinaria <strong>di</strong> acido t,t-muconico <strong>di</strong> altra origine: alimenti e bevande, conservanti<br />
presenti negli alimenti (acido sorbico), fumo <strong>di</strong> sigaretta, ecc. (3,<br />
4, 5). Alcuni autori hanno anche indagato i valori <strong>di</strong> escrezione urinaria<br />
<strong>di</strong> acido t,t-muconico urinario in popolazioni <strong>di</strong> soggetti non professionalmente<br />
esposti a benzene (Tab. I). Il valore BEI proposto dall’ACGIH<br />
per l’acido t,t-muconico urinario è <strong>di</strong> 500 µg/g <strong>di</strong> creatinina.
