E’ invece coinvolta l'attivazione della PI3K, visto che l'inibitore farmacologico della PI3K, l'LY 294002 ha bloccato l'effetto del 17β- estra<strong>di</strong>olo. Questo dato è in accordo con i risultati <strong>di</strong> altri laboratori, che vedono attività antinfiammatoria della fosfati<strong>di</strong>linositolo 3-chinasi [191- 195], ed interazioni tra ER e la subunità regolatoria della fosfati<strong>di</strong>linositolo 3-chinasi sono state descritte in più sistemi cellulari [131, 132]. Valutando questi risultati si può ipotizzare che l'azione dell'E2 sull'attivazione <strong>di</strong> p65 è un evento non genomico, in quanto si esplica in tempi troppo brevi (<strong>di</strong>eci minuti) per essere compatibili con una attivazione della espressione genica. È importante inoltre notare che il 17β-estra<strong>di</strong>olo blocca l'attivazione <strong>di</strong> p65 in modo in<strong>di</strong>pendente dalla degradazione <strong>di</strong> IκBα, <strong>di</strong>fferenziandosi in questo dai FANS, che invece agiscono inibendo IKK. Infine in questa tesi <strong>di</strong>mostriamo che l'azione inibitoria del 17β-estra<strong>di</strong>olo sull'attivazione <strong>di</strong> NF-κB si esplica solo nelle cellule macrofagiche. Ulteriori <strong>stu<strong>di</strong></strong> sono necessari per meglio caratterizzare gli interme<strong>di</strong> che, oltre alla fosfati<strong>di</strong>linositolo 3-chinasi, me<strong>di</strong>ano l'azione dell'estrogeno. Questi <strong>stu<strong>di</strong></strong> potranno servire per in<strong>di</strong>viduare dei nuovi bersagli nella via <strong>di</strong> attivazione <strong>di</strong> NF-κB che agiscono a monte della trascrizione <strong>di</strong> geni infiammatorii. Lo sviluppo <strong>di</strong> nuovi farmaci che agiscono in modo simile all'E2 potrà essere utile per prevenire l'insorgenza <strong>di</strong> quelle malattie dove la componente infiammatoria svolge un ruolo cruciale nella loro patogenesi. 122
