<strong>protéine</strong>s paxilline, p130Crk-associated substrate (Cas), <strong>et</strong> les <strong>protéine</strong>s kinases ERK<br />
(Extracellu<strong>la</strong>r Regu<strong>la</strong>ted Kinase) <strong>et</strong> MLCK (Myosin Light Chain Kinase) (Webb, Donais <strong>et</strong><br />
al. 2004; Zai<strong>de</strong>l-Bar, Milo <strong>et</strong> al. 2007). Puisque l’activation <strong>de</strong> RhoA est l’événement clé pour<br />
<strong>la</strong> formation <strong>de</strong>s adhérences focales, leur désassemb<strong>la</strong>ge va souvent nécessiter son<br />
inactivation. La FAK peut faciliter <strong>la</strong> dissociation <strong>de</strong>s adhérences formées en activant<br />
p190RhoGAP qui, en inhibant RhoA, diminue l’activité <strong>de</strong> ROCK. FAK recrute également<br />
aux sites d’adhérence un complexe contenant Rac1, PIX (un facteur d’échange <strong>de</strong> Cdc42 <strong>et</strong><br />
Rac) <strong>et</strong> PAK (un effecteur <strong>de</strong> Rac1), c<strong>et</strong>te <strong>de</strong>rnière kinase inactive <strong>la</strong> MLCK (San<strong>de</strong>rs,<br />
Matsumura <strong>et</strong> al. 1999) <strong>et</strong> réduit les fibres <strong>de</strong> stress (Schober, Raghavan <strong>et</strong> al. 2007). La<br />
paxilline est un substrat <strong>de</strong> FAK <strong>et</strong> <strong>la</strong> forme phosphorylée peut recruter soit le complexe<br />
CrkII-p130Cas-DOCK180 (Kiyokawa, Hashimoto <strong>et</strong> al. 1998; Kiyokawa, Hashimoto <strong>et</strong> al.<br />
1998), DOCK180 étant un facteur d’échange <strong>de</strong> Rac1, soit <strong>la</strong> <strong>protéine</strong> p120RasGAP<br />
(Tsubouchi, Sakakura <strong>et</strong> al. 2002) qui inhibe RhoA. Egalement, <strong>la</strong> <strong>protéine</strong> kinase dépendante<br />
du calcium <strong>et</strong> <strong>de</strong> <strong>la</strong> calmoduline <strong>de</strong> type II ou CaMKII perm<strong>et</strong>trait <strong>la</strong> motilité cellu<strong>la</strong>ire en<br />
stimu<strong>la</strong>nt localement <strong>et</strong> transitoirement <strong>la</strong> phosphory<strong>la</strong>tion sur tyrosine <strong>de</strong>s <strong>protéine</strong>s <strong>de</strong>s<br />
adhérences focales (Easley, Brown <strong>et</strong> al. 2008).<br />
Les phosphatases<br />
Les phosphatases SHP-2 (Src-homology-domain2-containing tyrosine phosphatase-2) <strong>et</strong> PTP-<br />
PEST, PTEN (PTP1B and phosphatase and tensin homologue <strong>de</strong>l<strong>et</strong>ed from chromosome ten)<br />
sont impliquées <strong>dans</strong> <strong>la</strong> déstabilisation <strong>de</strong>s adhérences. La perte <strong>de</strong> leur expression <strong>dans</strong> les<br />
fibrob<strong>la</strong>stes conduit à une réduction <strong>de</strong> <strong>la</strong> migration ainsi qu’une augmentation du nombre <strong>et</strong><br />
<strong>de</strong> <strong>la</strong> taille <strong>de</strong>s adhérences focales (Yu, Qu <strong>et</strong> al. 1998; Angers-Loustau, Cote <strong>et</strong> al. 1999). Ces<br />
phosphatases sont associées aux adhérences <strong>et</strong> contribuent à <strong>la</strong> <strong>régu<strong>la</strong>tion</strong> <strong>de</strong> <strong>la</strong> motilité<br />
cellu<strong>la</strong>ire (Arregui, Balsamo <strong>et</strong> al. 1998; Tamura, Gu <strong>et</strong> al. 1998; Garton and Tonks 1999).<br />
L’activité <strong>de</strong> <strong>la</strong> <strong>protéine</strong> phosphatase PP2A (Protein Phosphatase 2A) est également impliquée<br />
<strong>dans</strong> <strong>la</strong> déstabilisation <strong>de</strong>s adhérences (Vil<strong>la</strong>lobos Campos and Schonthal 2000). C<strong>et</strong>te<br />
phosphatase s’associe constitutivement avec l’intégrine αIIbβ3 <strong>dans</strong> les p<strong>la</strong>qu<strong>et</strong>tes circu<strong>la</strong>ntes<br />
<strong>et</strong> maintien c<strong>et</strong>te intégrine inactive. Sa perte d’expression accélère l’adhésion <strong>de</strong>s p<strong>la</strong>qu<strong>et</strong>tes<br />
sur fibrinogène (Gushiken, Patel <strong>et</strong> al. 2008). D’autre part, <strong>la</strong> phosphory<strong>la</strong>tion <strong>de</strong> <strong>la</strong> FAK sur<br />
<strong>la</strong> tyrosine 925 perm<strong>et</strong> le recrutement via <strong>la</strong> <strong>protéine</strong> adaptatrice Grb2 <strong>de</strong> <strong>la</strong> dynamine au<br />
niveau <strong>de</strong>s adhérences, un mécanisme à l’origine du désassemb<strong>la</strong>ge <strong>de</strong>s sites adhésifs<br />
(Ezratty, Partridge <strong>et</strong> al. 2005). La tyrosine phosphatase déphosphory<strong>la</strong>nt c<strong>et</strong>te tyrosine 925<br />
pourrait être <strong>la</strong> phosphatase PTP1B puisque sa surexpression favorise <strong>la</strong> stabilité <strong>de</strong>s<br />
adhérences focales <strong>et</strong> réduit le taux <strong>de</strong> migration <strong>de</strong>s cellules (Arregui, Balsamo <strong>et</strong> al. 1998;<br />
Liu, Sells <strong>et</strong> al. 1998).<br />
III.3.2.2. Le désassemb<strong>la</strong>ge <strong>de</strong>s adhérences par protéolyse<br />
Une autre voie <strong>de</strong> désassemb<strong>la</strong>ge <strong>de</strong>s adhérences utilise <strong>la</strong> protéolyse <strong>de</strong>s composants<br />
par <strong>la</strong> calpaïne ou le protéasome. L’inhibition <strong>de</strong> <strong>la</strong> calpaïne, une protéase dépendante du<br />
calcium, conduit à <strong>de</strong>s adhérences focales plus <strong>la</strong>rges distribuées en périphérie <strong>de</strong> <strong>la</strong> cellule <strong>et</strong><br />
à une diminution <strong>de</strong> <strong>la</strong> motilité. L’organisation du cytosquel<strong>et</strong>te est aussi modifiée<br />
(Huttenlocher, Palecek <strong>et</strong> al. 1997; Dourdin, Bhatt <strong>et</strong> al. 2001). La calpaïne pourrait fragiliser<br />
les adhérences focales <strong>et</strong> perm<strong>et</strong>tre ainsi aux intégrines <strong>de</strong> se détacher du substrat. En eff<strong>et</strong>, le<br />
clivage <strong>de</strong> <strong>la</strong> taline par <strong>la</strong> calpaïne stimule <strong>la</strong> dissociation <strong>de</strong> plusieurs molécules présentes<br />
<strong>dans</strong> les adhérences dont <strong>la</strong> paxilline, <strong>la</strong> vinculine <strong>et</strong> <strong>la</strong> zyxine (Franco, Rodgers <strong>et</strong> al. 2004).<br />
Cependant, <strong>la</strong> tête <strong>de</strong> <strong>la</strong> taline issue du clivage par <strong>la</strong> calpaïne stimule toujours l’activation <strong>de</strong>s<br />
intégrines (Cal<strong>de</strong>rwood, Zent <strong>et</strong> al. 1999; Cal<strong>de</strong>rwood, Yan <strong>et</strong> al. 2002; Vinogradova, Velyvis<br />
