II.1.4. L’activité régu<strong>la</strong>trice <strong>de</strong> <strong>la</strong> CaMKII sur l’adhérence cellu<strong>la</strong>ire requiert <strong>ICAP</strong>-1α<br />
<strong>ICAP</strong>-1α est une phospho<strong>protéine</strong> dont le niveau <strong>de</strong> phosphory<strong>la</strong>tion augmente au cours <strong>de</strong><br />
l’adhérence <strong>de</strong>s cellules sur fibronectine (Chang, Wong <strong>et</strong> al. 1997; Zhang, Hemler <strong>et</strong> al.<br />
1999). Elle possè<strong>de</strong> <strong>dans</strong> sa région N-terminale un site consensus <strong>de</strong> phosphory<strong>la</strong>tion <strong>de</strong> <strong>la</strong><br />
CaMKII contenant une thréonine en position 38 potentiellement phosphorylée par <strong>la</strong> CaMKII.<br />
L’inhibition <strong>de</strong> l’intégrine par <strong>la</strong> CaMKII pourrait se faire par l’intermédiaire d’une <strong>protéine</strong><br />
régu<strong>la</strong>trice <strong>et</strong> <strong>ICAP</strong>-1α constitue à c<strong>et</strong> égard un bon candidat. En eff<strong>et</strong>, alors que <strong>la</strong><br />
substitution <strong>de</strong> <strong>la</strong> thréonine 38 d’<strong>ICAP</strong>-1α par une a<strong>la</strong>nine non phosphorylée (T38A) stimule<br />
très fortement l’étalement <strong>de</strong>s cellules CHO sur une matrice <strong>de</strong> fibronectine, sa mutation<br />
phosphomimétique en aspartate (T38D) réduit fortement les capacités adhésives <strong>de</strong>s cellules<br />
(Bouvard and Block 1998). La phosphory<strong>la</strong>tion d’<strong>ICAP</strong>-1α au niveau <strong>de</strong> <strong>la</strong> thréonine 38<br />
semble impliquée <strong>dans</strong> <strong>la</strong> <strong>régu<strong>la</strong>tion</strong> négative <strong>de</strong> l’étalement cellu<strong>la</strong>ire médiée par l’intégrine<br />
α5β1. Plus précisément, <strong>la</strong> mutation T38A stimule l’étalement cellu<strong>la</strong>ire <strong>de</strong> façon simi<strong>la</strong>ire à<br />
l’inhibition <strong>de</strong> <strong>la</strong> CaMKII par le KN-62 alors que <strong>la</strong> mutation T38D réduit l’adhérence <strong>de</strong>s<br />
cellules, comme l’activation constitutive <strong>de</strong> <strong>la</strong> CaMKII. L’expression <strong>de</strong>s mutants T38A ou<br />
T38D rend les cellules CHO insensibles à l’action d’un inhibiteur pharmacologique <strong>de</strong> <strong>la</strong><br />
CaMKII (KN-62). Ainsi <strong>la</strong> CaMKII est impliquée <strong>dans</strong> <strong>la</strong> phosphory<strong>la</strong>tion d’<strong>ICAP</strong>-1α<br />
puisque l’eff<strong>et</strong> <strong>de</strong>s mutations T38A <strong>et</strong> T38D sur l’adhérence cellu<strong>la</strong>ire est indépendant <strong>de</strong><br />
l’activité CaMKII. Ainsi, <strong>ICAP</strong>-1α étant un régu<strong>la</strong>teur négatif <strong>de</strong> l’intégrine β1, sa<br />
phosphory<strong>la</strong>tion pourrait contrôler son activité. Les données structurales suggèrent que <strong>la</strong><br />
région N-terminale d’<strong>ICAP</strong>-1α puisse masquer le site d’interaction à <strong>la</strong> sous-unité β1. Il est<br />
difficile <strong>de</strong> co-immunoprécipiter les <strong>de</strong>ux <strong>protéine</strong>s sans avoir recours à <strong>de</strong>s fusions qui<br />
changent vraisemb<strong>la</strong>blement <strong>la</strong> conformation d’<strong>ICAP</strong>-1α. <strong>ICAP</strong>-1α pourrait être régulée par<br />
une phosphory<strong>la</strong>tion qui déplie sa structure <strong>et</strong> expose le domaine PTB d’interaction avec<br />
l’intégrine. Cependant, <strong>la</strong> phosphory<strong>la</strong>tion in vivo d’<strong>ICAP</strong>-1α par <strong>la</strong> CaMKII n’a pas encore<br />
été démontrée.<br />
D’autres sites consensus <strong>de</strong> phosphory<strong>la</strong>tion ont été i<strong>de</strong>ntifiés <strong>dans</strong> <strong>la</strong> <strong>protéine</strong> <strong>ICAP</strong>-1α,<br />
principalement localisés <strong>dans</strong> sa partie N-terminale : trois sites PKC <strong>et</strong> un site PKA. La<br />
fonction biologique <strong>de</strong> ces phosphory<strong>la</strong>tions n’a pas encore été mise en évi<strong>de</strong>nce. Cependant,<br />
<strong>la</strong> PKC ne semble pas avoir d’action directe <strong>dans</strong> le contrôle <strong>de</strong> l’affinité <strong>de</strong> l’intégrine α5β1<br />
(Bouvard, Mol<strong>la</strong> <strong>et</strong> al. 1998). Elle semblerait plutôt impliquée <strong>dans</strong> le recyc<strong>la</strong>ge <strong>de</strong>s intégrines<br />
à <strong>la</strong> membrane p<strong>la</strong>smique (Ng, Shima <strong>et</strong> al. 1999).<br />
