phospholipides et des différentes classes de lipides constitutifs. Il serait aussi possible de réaliser un co-immunomarquage de la protéine PLD1 et de marqueurs spécifiques des microdomaines tels que le GM1. Ceci permettrait de voir l’effet des régimes sur la localisation de la PLD dans les radeaux lipidiques par électrophorèse et par des approches en microscopie à fluorescence. Les anticorps anti-PLD1 et PLD2 dont nous disposons pourraient être utilisés dans des expériences d’immunofluorescence après perméabilisation des cellules, couplage à un anticorps secondaire et fixation des thymocytes sur lames. Cette approche permettrait de déterminer s’il existe une relation entre l’enrichissement en acides gras des radeaux lipidiques et la délocalisation de la protéine PLD. Pour compléter cette étude, il serait intéressant d’étudier les effets des régimes sur la composition en acides gras et l’activité PLD d’autres organes (rate, foie, cœur, tissu adipeux). Il serait aussi important de confirmer nos résultats obtenus chez le rat par une étude clinique. Cette étude pourrait prendre en compte des paramètres supplémentaires fonctionnels comme la production de cytokines ou l’activité des cellules NK ou, biochimiques comme la production d’eicosanoïdes et l’expression des molécules de surface. 162
