A similar mechanism can be postulated for hemocyanin, with the occurrence of a dynamic heterogeneity influenced by the presence of effectors such as oxygen and lactate, resulting in a stabilized asymmetric binding site when L-lactate is effectively bound. Performing studies similar to those made on hemoglobin and DPG with hemocyanin and L-lactate would help to test this hypothesis. It has been showed that the oxygenated Hc hexamer of Panulirus interruptus has a reduced channel along the three-fold axis compared to the deoxygenated form (Haas et al., 1993). Using the simple Monod-Wyman-Chnageux (MWC) model for Carcinus maenas Hc dodecamer, Weber and collaborators showed that L-lactate increases O2 affinity in part by shifting the allosteric equilibrium towards the high-affinity R state (Weber et al., 2008). From these data it can be suggested that the potential binding of lactate in the reduced central channel could stabilize the oxygenated R conformation. Such a hypothesis must be considered with care and at the hexamer scale, since SAXS studies of the Hc dodecamer of Homarus americanus have showed that the oxygenated dodecamer exists in two different forms, the one without lactate and the one in the presence of lactate with the two hexamers closer by 0.5 nm. In this case the simple MWC model is not longer relevant and nested allostery must be considered (Hartmann et al., 2001). It would also be of interest to perform studies similar to those conducted here using hemocyanin from terrestrial crabs or mud shrimps with little or no lactate effect, and to test the effect or other effectors such as urate or D-lactate. BILAN ET PERSPECTIVES 131 3.3 Bilan et perspectives Les études présentées dans ce chapitre ont permis de mettre en évidence l’interaction spécifique du L-lactate avec les deux sous-unités les plus légères de l’Hc de Carcinus maenas. La liaison du L- lactate sur chacune de ces sous-unités isolées n’a pas été observée ; le site de fixation est plutôt formé par la structure quaternaire de l’Hc, au niveau de l’axe de symétrie d’ordre 3 des hexamères. Ainsi la fixation du L-lactate à l’Hc de Carcinus maenas fait intervenir à la fois des sous-unités spécifiques avec lesquelles il interagit et la structure quaternaire globale du complexe. Le rôle central des cations divalents dans l’assemblage des dodécamères a également été mis en évidence : l’incorporation de la sous-unité spécifique du dodécamère est inhibée par la chélation des ions divalents et l’assemblage est essentiellement limité à la forme hexamèrique. Les interactions entre les sous-unités et les cations divalents impliquées dans la formation des hexamères sont différentes de celles impliquées dans la formation des dodécamères. Ces résultats fournissent des informations sur les modalités de l’interaction entre le L-lactate et l’hémocyanine mais ne permettent pas de déterminer les acides aminés formant le site actif ni les changements structuraux provoqués par la fixation du L-lactate. Ces données seraient intéressantes pour comprendre de quelle manière la liaison du L-lactate augmente l’affinité du pigment pour l’oxygène. La cristallisation d’un complexe d’Hc en présence de L-lactate pourrait donner des informations sur la localisation précise du site actif mais peut se révéler difficile à mettre en oeuvre. Une approche en cryomicroscopie électronique pourrait permettre de reconstruire le volume du complexe en l’absence et en présence de L-lactate et d’observer les changements résultant de la fixation du L-lactate. Le chapitre suivant présente des travaux réalisés selon cette méthode.
