Nous étudierons également les interactions des cellules cancéreuses avec les cellules du système immunitaire et notamment les macrophages. Il a en effet été montré que les macrophages facilitent l’invasion du stroma et des vaisseaux sanguins par les cellules de cancer du sein, via un mécanisme de chimiotaxie [Robinson 09, Patsialou09, Erez10]. Finalement, nous chercherons à recréer au moins un vaisseau sanguin dans les matrices artificielles afin d’étudier comment les cellules cancéreuses interagissent avec les cellules endothéliales des vaisseaux. Il a en effet été montré que l’on peut construire un vaisseau sanguin in vitro avec 3 types de cellules (cellules de muscle lisse, cellules endothéliales et fibroblastes), du collagène, un milieu nutritif adapté et des pièces mécaniques spécifiques, en forme d’essieu [L’Heureux93, Isenberg06]. Le développement d’un modèle biologique de matrice humaine, en trois dimensions, très proche de l’in vivo pour la croissance tumorale est le principal objectif et résultat attendu. Ce modèle humain pourra être ensuite utilisé pour de nombreuses applications: - Etude des interactions entre les différents constituants de la matrice avec les cellules tumorales et estimation de l’importance de chacun. - Tests de médicaments, étude de l’efficacité de traitements.
