ABSTRACT<br />

PRELIMINARY IN VITRO SCREENING USING A PERA DISTICHOPHYLLA ALCOHOLIC<br />

EXTRACT (EC) AGAINST AN ACANTHAMOEBA SPP ISOLATE RESPONSIBLE OF<br />

CORNEAL ULCER<br />

Actually, infections due to Acanthamoeba spp have increased, specially at cornea and its treatment has<br />

limitations. New options for treatment have been proved, including a large number of therapeutic agents and<br />

natural products, which have been tested in vitro to evaluate the amoebicidal activities against this pathogen<br />

organism. In this sense, we proposed to evaluate in vitro biological activities of cru<strong>de</strong> alcoholic extract (Ec)<br />

from <strong>Pera</strong> <strong>distichophylla</strong>, an amazonic plant, against an Acanthamoeba spp isolate (A27), obtained from<br />

a patient who suffered a corneal ulcer. We evaluated the effects of 1 and 10 mg/ml concentrations of Ec<br />

against Acanthamoeba spp trophozoites and cysts in 24-wells polystyrene microplates. Growth amoeba<br />

inhibition was observed, at both Ec concentrations exposure, being greater at 10 mg/ml; morphologic<br />

changes were also <strong>de</strong>tected in vegetative forms, as ro<strong>un</strong>ding and vacuolization, signs of cell <strong>de</strong>generation<br />

or <strong>de</strong>ath. No morphologic variations at light microscopy were seen in cystic forms after exposure to both<br />

Ec concentrations. These results suggest that Ec could produce trophozoites lysis and have amoebostatic,<br />

lytic and amoebicidal effects at 1 and 10 mg/ml concentrations. We recommend to complement this study<br />

evaluating other concentrations of Ec and possible ultrastructural cell changes by other techniques like<br />

electronic microscopy.<br />
