ABSTRACT<br />

BIOLOGICAL ACTIVITY OF HEXANOLIC (EH) AND METHANOLIC (EM) EXTRACTS<br />

FROM PERA DISTICHOPHYLLA AGAINST AN ACANTHAMOEBA SPP. ISOLATE<br />

OBTAINED FROM A CORNEAL ULCER<br />

Recently, infections due to Acanthamoeba genus have increased, specially ocular affections and<br />

pharmacological treatment for them has some limitations. Searching for effective alternatives against<br />

this protozoan, we <strong>de</strong>ci<strong>de</strong>d to evaluate natural products obtained from plants, having previous reports of<br />

amoebicidal activity of some extracts against different species of Acanthamoeba. We evaluated in vitro<br />

biological activity of hexanolic (<strong>Eh</strong>) and methanolic (<strong>Em</strong>) extract, obtained from <strong>Pera</strong> <strong>distichophylla</strong>, an<br />

amazonic plant, against an Acanthamoeba spp. (A27) strain, isolated from a patient suffering a corneal<br />

ulcer. Trophozoites of isolate A27 exposed to different <strong>Eh</strong> concentrations presented morphological changes,<br />

probably caused by the effect of DMSO as solvent extract, while morphological changes observed after<br />

contact with <strong>Em</strong> seemed to be caused by 0.75 mg/ml <strong>Em</strong> concentration and not by the DMSO; no changes<br />

were observed in the cysts. The 0.75 mg/ml <strong>Eh</strong> concentration apparently <strong>de</strong>creased the proliferation<br />

of Acanthamoeba spp. and <strong>Em</strong> produced lysis. Viability tests not yiel<strong>de</strong>d important changes. Finally,<br />

we recommend evaluating these extracts on other Acanthamoeba spp. isolates, to <strong>de</strong>termine whether<br />

these effects are isolate-<strong>de</strong>pen<strong>de</strong>nt; performing cytotoxicity tests on corneal cells and in vivo studies in<br />

experimental animals, which would provi<strong>de</strong> more information about its activity and prophylactic potential<br />

or therapeutic use.<br />
