ABSTRACT
BIOLOGICAL ACTIVITY OF HEXANOLIC (EH) AND METHANOLIC (EM) EXTRACTS
FROM PERA DISTICHOPHYLLA AGAINST AN ACANTHAMOEBA SPP. ISOLATE
OBTAINED FROM A CORNEAL ULCER
Recently, infections due to Acanthamoeba genus have increased, specially ocular affections and
pharmacological treatment for them has some limitations. Searching for effective alternatives against
this protozoan, we decided to evaluate natural products obtained from plants, having previous reports of
amoebicidal activity of some extracts against different species of Acanthamoeba. We evaluated in vitro
biological activity of hexanolic (Eh) and methanolic (Em) extract, obtained from Pera distichophylla, an
amazonic plant, against an Acanthamoeba spp. (A27) strain, isolated from a patient suffering a corneal
ulcer. Trophozoites of isolate A27 exposed to different Eh concentrations presented morphological changes,
probably caused by the effect of DMSO as solvent extract, while morphological changes observed after
contact with Em seemed to be caused by 0.75 mg/ml Em concentration and not by the DMSO; no changes
were observed in the cysts. The 0.75 mg/ml Eh concentration apparently decreased the proliferation
of Acanthamoeba spp. and Em produced lysis. Viability tests not yielded important changes. Finally,
we recommend evaluating these extracts on other Acanthamoeba spp. isolates, to determine whether
these effects are isolate-dependent; performing cytotoxicity tests on corneal cells and in vivo studies in
experimental animals, which would provide more information about its activity and prophylactic potential
or therapeutic use.
