ABSTRACT<br />

IN VITRO SUSCEPTIBILITY TO NIFURTIMOX AND BENzNIDAzOLE TO TRYPANOSOMA<br />

CRUZI ISOLATES OBTAINED FROM VENEzUELAN PATIENTS WITH CHAGAS DISEASE<br />

INFECTED BY ORAL AND VECTOR TRANSMISSION MECHANISMS)<br />

The frequent reports of outbreaks of acute Chagas disease (CD) by ingestion of contaminated food,<br />

illustrate the importance of this transmission mo<strong>de</strong>l. The present study, evaluate for the first time the in vitro<br />

susceptibility of parasite isolates obtained from Venezuelan patients with Chagas’ disease infected by oral<br />

and vector transmission mechanisms to the drugs Nifurtimox and Benznidazole. We used epimastigotes<br />

and trypomastigotes of Trypanosoma cruzi isolates from four patients with CD. Concentrations of 1x10 8<br />

P/mL were incubated for 72 hours with drugs Nifurtimox and Benznidazole in a range of 0.03 to 4000μM.<br />

Used the MTT colorimetric technique for assessing cell viability and calculated the IC 50 values for each<br />

of the isolated parasites. The isolates tested and two parasitic <strong>de</strong>velopmental stages were shown to be<br />

susceptible to Nifurtimox and Benznidazole drugs with IC 50 values vary greatly among themselves. The<br />

statistical analysis showed that the drug Nifurtimox was more effective than Benznidazole (p < 0.05) in the<br />

trypomastigote stage and two stage epimastigote isolates. We could characterize the in vitro activity of the<br />

drugs Nifurtimox and Benznidazole against human isolates of T. cruzi with different routes of transmission<br />

in Venezuela, showing greater sensitivity on the epimastigote stage with respect to the trypomastigote stage.<br />
