Calle 110 no. 514 e/ 5. a B y 5. a F, Playa, Ciudadde La Habana, CP 11600, ybaro@inisav.cuRESUMENSe analizó por cromatografía líquida de alta resolución (HPLC) lapresencia de β-exotoxina en nueve cepas de Bacillus thuringiensis,pertenecientes a la colección del Instituto de Investigaciones deSanidad Vegetal. La concentración de β-exotoxina en el patrón fuede 25 µg.mL –1 , y en el producto comercial empleado como controlpositivo fue de 6 µg.mL –1 . En las cepas evaluadas solo la LBT9 yLBT47 mostraron la presencia de este metabolito a concentracionesde 34 y 4,5 µg.mL –1 respectivamente.Palabras claves: Bacillus thuringiensis, β-exotoxina, cromatografíalíquida (HPLC), detecciónABSTRACTThe presence of β-exotoxin in nine Bacillus thuringiensis strainsbelonging to Plant Health Research Institute collection was analyzedby high performance liquid chromatography (HPLC). β-exotoxinconcentration in the standard was 25 µg.mL -1 , and in a commercialproduct, used as positive control, was 6 µg.mL -1 . β-exotoxin was onlydetected in LBT9 with 34 µg.mL -1 and LBT47 with 4.5 µg.mL -1 , amongthe strains proved.Key words: Bacillus thuringiensis, β-exotoxin, liquid chromatography(HPLC), detectionINTRODUCCIÓNAlgunas cepas de Bacillus thuringiensis producen ademásde la δ-endotoxinas o proteínas Cry una exotoxinatermoestable denominada β-exotoxina, la cual se secretaal medio de cultivo al inicio del proceso deesporulación. Esta molécula es un análogo nucleotídicode ADN con un peso molecular de 701 Da y se le atribuyeacción insecticida contra diferentes órdenes deinsectos como Lepidópteros, Dípteros, Coleópteros,Hemípteros, además de ácaros, nematodos, protozoariosy platelmintos [Hernández et al., 2003].Diversos métodos como la cromatografía de intercambioiónico, electroforesis capilar, ELISA y la cromatografíalíquida de alta resolución (HPLC) se han descritopara la determinación de la β-exotoxina, comoalternativas al bioensayo tradicional con Musca domestica,el cual resulta más trabajoso, prolongado en eltiempo y presenta algunas limitaciones como estimacionesinexactas de la potencia de la toxina impura ypoca reproducibilidad. De todos estos métodos la RP-HPLC se emplea más frecuentemente para la detección ycuantificación de este metabolito [Gohar y Perchat, 2001].El presente trabajo tuvo como objetivo detectar y cuantificarla presencia de la β-exotoxina en nueve cepascubanas de Bacillus thuringiensis.MATERIALES Y MÉTODOSComo material biológico se emplearon nueve cepas deB. thuringiensis pertenecientes a la colección del Institutode Investigaciones de Sanidad Vegetal, las cualesse seleccionaron de acuerdo con su serotipo flagelar, y latoxicidad exhibida en el sobrenadante del cultivo esterilizadoa 121ºC frente a un grupo de organismos plaga(Tabla 1).El estándar de β-exotoxina I lo suministró el doctor JorgeIbarra (Laboratorio de Bioinsecticidas del Centro defitosanidad/255
