3. As the swab is withdrawn from the flask, carefully press it<br />
into the agar to make a deep cut in the center of the inoculated<br />
area. The swab will not make a clean cut; however,<br />
a jagged cut will not adversely affect the performance of<br />
Mycotrim <strong>RS</strong>.<br />

3. Incubation<br />
Incubate the flask agar side up at 34° C to 37° C for 72 hours<br />
or until the color of the media changes. Check the flask for a<br />
color change every 24 hours (see 4. Observation, below).<br />
Temperatures above 38° C will inhibit M. pneumoniae.<br />
When removing flasks from the incubator for examination,<br />
care should be taken to avoid washing growth off the agar by<br />
splashing the broth over the agar.<br />
4. Observation<br />
A. Color change<br />
The first sign of M. pneumoniae in Mycotrim <strong>RS</strong> is a color<br />
change in the media which may occur within 72 to 96 hours<br />
after inoculation. Growth of M. pneumoniae will cause the<br />
media to change from red to yellow-orange.<br />
Unlike bacteria and fungi, M. pneumoniae does not cause<br />
turbidity in the broth, even at maximum titers. Gross turbidity<br />
of visible colonies on the agar indicate bacterial or fungal<br />
overgrowth, making the color change unreliable and possibly<br />
hiding mycoplasma colonies. If no color change occurs<br />
within 72 hours, reinoculate the agar as indicated in step 5.<br />
