DIRECTIONS FOR USE<br />

1. Preparation of Flask<br />

A. Loosen the flask cap 1/4 to 1/2 turn and warm the flask<br />

in the incubator (34° C to 37° C) for approximately five<br />

minutes.<br />

B. Aseptically add antimicrobial discs according to specimen<br />

type:<br />

Specimen Source Antimicrobial Discs<br />

Throat 1 CN, 1 TA<br />

Bronchial washings 1 CN, 1 TA<br />

Pneumocentesis fluid 1 CN, 1 TA<br />

Tissue from biopsy 1 CN, 1 TA<br />

Sputum 2 CN, 2 TA<br />

C. Recap the flask tightly and place it agar side down, as<br />

shown below in Figure 1, for a minimum of 30 minutes.<br />

2. Inoculation<br />

Use aseptic technique.<br />

A. For liquid specimens such as bronchial washings, pneumocentesis<br />

fluid and washings of tissue biopsy:<br />

1. Hold the flask at about a 45° angle as shown in Figure 2<br />

and allow approximately 1.0 mL of specimen to slow flow<br />

down the center of the agar.<br />

2. As the pipet is withdrawn from the flask, press it firmly into<br />

the agar to make a deep cut in the area inoculated.<br />
