Objetivos da unidade curricular e competências a desenvolver (1000 caracteres);
Definir e caracterizar anticorpo e antigénio;
Descrever o modo como o anticorpo e o antigénio interagem em Imunohistoquímica (IHQ);
Identificar, interpretar e criticar os vários métodos e técnicas em IHQ;
Reconhecer e interelacionar os diversos fatores que influenciam os métodos e técnicas;
Indicar as diferentes aplicações da IHQ e a sua importância como método complementar de diagnóstico;
Planear e executar diferentes técnicas de IHQ assim como testar e aferir anticorpos.

Define and characterize antibody and antigen;
Describe how the antibody and antigen interact in Immunohistochemistry (IHC);
Identify, interpret and criticize the various methods and techniques in IHC);
Recognize the diverse and interrelated factors that influence the methods and techniques;
Indicate the several applications of IHC and its importance as a complementary method of
diagnosis;
Plan and implement different techniques for IHC as well as test and measuring antibodies.
Conteúdos programáticos (1000 caracteres):
Fundamentos da imunohistoquímica.
Anticorpos específicos: monoclonais vs policlonais; sensibilidade e especificidade; critérios de
especificidade.
Sistemas de deteção : direto , indireto , enzimas solúveis imunocomplexas, ABC, LAB, LSAB, polímero,
entre outros.

Técnicas de imunohistoquímica: método de fluorescência direta e indireta , ABC, LSAB e APAAP.
Técnica de LSAB aplicada ao músculo.
Recuperação antigénica.
Bloqueio de substâncias endógenas, de radicais livres e permeabilização das membranas.
Diluição de anticorpos.
Fixação: diferentes tipos de fixadores, vantagens e desvantagens.
Descalcificação: implicações em imunohistoquimica.
Controlo de qualidade interno e externo.
Painéis de anticorpos mais usuais em IHQ e suas aplicações.
Preparação, planeamento e execução de técnicas IHQ.

Fundamentals of immunohistochemistry.
Specific antibodies: monoclonal vs polyclonal; sensitivity and specificity, criteria for specificity.
Detection systems: direct, indirect, enzyme soluble immune complexes, ABC, LAB, LSAB, polymer,
among others.
Immunohistochemical techniques: method of direct and indirect fluorescence, ABC, LSAB, and
APAAP.
LSAB technique applied to the muscle.
Recovery antigen.
Blocking endogenous substances, free radicals and membrane permeabilization.
Dilution of antibodies.
Fixation: different types of fixatives, advantages and disadvantages.
Decalcification: implications for immunohistochemistry.
Internal and external quality control.
Panels of antibodies more usual in IHC and its applications.
Preparation, planning and execution of IHC techniques.
