bARLEy, MALT AND bEER<br />

Introduction<br />

Strobilurin pesticides represent an important group of<br />

fungicides with a broad efficiency spectrum. These fungicides<br />

began to be used commercially in 1996 and they were<br />

produced by the companies Syngenta (azoxystrobin) and<br />

BASF (kresoxim-methyl) 1 .<br />

natural strobilurins (Strobilurus and Oudemansiella)<br />

were isolated from wood-rotting bracket fungi (Basidiomycetes).<br />

These wood decay fungi produce fungicidially<br />

acting substances in defense against lower fungi. The name<br />

strobilurine comes from Strobilurus tanacetus, one of the first<br />

fungi from which they were isolated (strobilurin A) 1 .<br />

Strobilurins represent an important group of efficient<br />

fungicides and together with oxazoline-diones (famoxadone)<br />

and imidazolinones (fenamidone) they are Qo inhibitors<br />

(Quinone outside Inhibitors). Their action is based on inhibition<br />

of mitochondrial respiration, they bind Qo instead of<br />

cytochrom b, which is a part of cytochromal complex bc1<br />

localized in the inner mitochondrial membrane of fungi.<br />

Binding blocks the transfer of electrons between cytochrom<br />

b and cytochrom c1, inhibiting thus production of ATP and<br />

disrupting the energy cycle of fungi. Originally, strobilurins<br />

are natural substances produced by higher fungi, e.g. Strobilurus<br />

tanacetus (strobilurin A), Oudimansiella mucida<br />

(oudemansin A). Chemically they belong to methoxyacrylates<br />

(e.g. azoxystrobin and picoxystrobin), methoxycarbamates<br />

(pyraclostrobin), oximino acetates (kresoxim – methyl,<br />

trifloxystrobin), ozimino-acetamides (dimoxystrobin, etc.),<br />

and dihydro-dioxazinones (fluoxastrobin). Their characters,<br />

including efficiency spectrum, are very variable. They affect<br />

mainly preventively, but also curatively and some even have<br />

eradicative effects. All of them act in contact, some of them<br />

in depth or translaminarily or systemically (azoxystrobin<br />

and picoxystrobin). They move acropetally in a plant. Their<br />

efficacy is very wide, they are active against sac fungi (Ascomycetes),<br />

bracket fungi, imperfect fungi and oomycetes.<br />

They are highly imperiled by the origin of resistance (cross<br />

resistance within the QoI fungicides) 1,2 .<br />

Resistance was proved in more pathogenic fungi. In<br />

cereals, powdery mildew (Blumeria graminis f.sp. tritici,<br />

Blumeria graminis f.sp. hordei), septoria blotch (Mycosphaerella<br />

graminicola), net blotch (Pyrenophora teres),<br />

helminthosporioses in wheat (Pyrenophora tritici – repentis),<br />

and in the other crops, e.g. in apple, scab (Venturia<br />

inaequalis), grapevine downy mildew (Plasmopara viticola),<br />

and grapevine powdery mildew (Erysiphe necator). When<br />

applying strobilurins, observance of all provisions is highly<br />

recommended to avoid the development of resistance 1,2,3 .<br />

Following fungicides are used against fungal diseases<br />

of barley in the Czech Republic azoxystrobin (Amistar),<br />

picoxystrobin (Acanto, Acanto Prima), trifloxystrobin (Sfera<br />

267.5 EC), fluoxystrobin (Fandango 200 EC), and kresoximmethyl<br />

(Juwel, Juwel Top). Azoxystrobin (Ortiva) is registered<br />

against fungal diseases of hop in the CR.<br />

Considering the increase in the extent of strobilurin use<br />

in the protection of barley and hop and their stability in a<br />

plant, it is necessary to introduce monitoring of their residues,<br />

intermediaries and final product.<br />

Experimental<br />

25 samples of beer, 50 samples of malt and 50 samples<br />

of barley were analyzed.<br />

60 ml of methanol/acetone mixture was added to 20 g of<br />

ground matrix. Disintegration of the mixture was performed<br />

in an ultrasound bath for 30 minutes. The disintegrated<br />

mixture was centrifuged at 6,500 rev min –1 for 15 min at<br />

10–15 °C. The obtained solutions were transferred to boiling<br />

flasks (250 ml) and evaporated to dryness on a vacuum evaporator.<br />

The obtained dry residue was dissolved with 20 ml<br />

of distilled water in the ultrasound bath for 1 minute. Mixture<br />

was purified through the SPE column EnVICarbII/PSA.<br />

pH of the beer sample (100 ml) is adjusted to pH 6 by<br />

adding naOH solution and the sample is then purified through<br />

the LiChrolut column.<br />

The analyses of samples were performed on a gas chromatograph<br />

(Trace GC Ultra, Thermo Finnigan) connected<br />

to a mass detector (Trace DSQ, Thermo Finnigan). To separate<br />

the analyzed substances a DB5-MS capillary column<br />

(30 m × 0.25 mm i.d., 0.25 mm) with following thermal program<br />

was used: initial temperature 70 °C for 1 min., increase<br />

in temperature 10 °C min –1 to 280 °C, maintained for 5 min.<br />
