The Metra <strong>CICP</strong> <strong>EIA</strong> kit provides a quantitative measure of the C-<strong>terminal</strong><br />

propeptide of type I collagen in serum. Collagen is synthesized as<br />

procollagen. C- and N-<strong>terminal</strong> peptides are cleaved from the collagen<br />

molecule prior to incorporation into the bone matrix and are released<br />

into the circulation.<br />

Elevated levels have been observed in diseases which are associated<br />

with high bone turnover.<br />

SAMPLE PREPARATION<br />

Sample type: Serum, cell culture medium<br />

Avoid haemolysis of specimens. Keep samples refrigerated at 2–8° C for<br />

storage of less than 5 days. For longer storage freeze specimens at<br />

-20° C or below.<br />

ASSAY PROCEDURE<br />

Dilute samples 1:12 with Assay Buffer Solution<br />

Label frame with the required number of Assay Strips<br />

Pipette 100 µl Standards, Controls, and diluted samples<br />

Incubate 120 ± 5 minutes at 18 – 25° C.<br />

Dissolve Enzyme Conjugate in<br />

Wash Buffer, use within 2 h<br />

Wash 3 times with 300 µl Wash Buffer<br />

Add 50 µl Stop Solution.<br />

Dilute Wash Buffer Concentrate<br />

1:10 with dist. water<br />

Add 100 µl Anti-<strong>CICP</strong> to each well<br />

Incubate 45 - 50 minutes at 18 – 25°C<br />

Add 100 µl Enzyme Conjugate to each well.<br />

Incubate 45 - 50 minutes at 18 – 25°C<br />

Prepare Substrate Solution 30-<br />

60 minutes prior to use. Allow<br />

tablets to dissolve completely.<br />

Wash 3 times with 300 µl Wash Buffer<br />

Wash 3 times with 300 µl Wash Buffer<br />

Add 100 µl Substrate Solution to each well<br />

Incubate 30 - 35 minutes at 18 – 25°C<br />

Read Optical Density at 405 nm<br />

(Reference measurement should be performed at 595 – 650 nm.)<br />

Analyze the assay results<br />

(Quantification software, 4-parameter fit: y = (A-D)/(1+(x/C)^B)+D )<br />
