IMPORTANCE, AIMS AND POSSIBLE OUTCOME OF RESEARCH<br />

My research project is focused on the in vitro effects of methane on xanthine<br />

oxidoreductase (XOR) enzyme and peroxynitrite (PN). XOR is the main source<br />

of reactive oxygen species (ROS) in ischaemia/reperfusion (IR), while in hypoxic<br />

circumstances it can play a role nitric oxide (NO) production due to its nitrite and<br />

nitrate reductase activity. NO and superoxide can form PN which causes lipid<br />

peroxidation and protein nitration. It has been shown that exogenic methane<br />

decreases the oxidative and nitrosative stress. Our aim was to find the mechanisms<br />

though which methane can influence inflammatory processes. Our hypotheses<br />

on one hand were that methane due to its protein conformation modifying<br />

activity can influence the enzyme activity of XOR, and on the other hand it can<br />

decrease nitrosative stress by reacting with PN. In our present studies we use<br />

flourometry for measuring the kinetic activity of XOR, chemilouminometry for<br />

measuring the amount of ROS produced by XOR, photometry for measuring the<br />

changes in nitrate/nitrite levels and enzyme-linked immunosorbent assay (ELI-<br />

SA) for measuring the amount of PN. In the future we would like to use gas chromatography-mass<br />

spectrometry (GC-MS) and NO trapping methods to precise<br />

the exact reaction between methane and PN.<br />

AIMS DURING HER CAREER<br />

I hope my research career will lead to a PhD degree and I do hope that I can witness<br />

that methane is used in clinics as a bioactive gas in decreasing IR injuries.<br />
