Stage 6: Quantitation and Normalization<br />

ABOUT THIS STAGE<br />

During this stage, you will prepare three independent dilutions of<br />

each PCR product in optical plates. You will then quantitate the<br />

diluted PCR products and average the OD measurements from each<br />

plate.<br />

Once the concentration of each reaction is determined, you will<br />

normalize each reaction to 2 µg/µL in RB Buffer.<br />

- Quantitation: 1 hour<br />

- Normalization: 1 hour<br />

Input required from Stage 5: PCR Product Purification and Elution is:<br />

Refer to Appendix A, Reagents, Equipment, and Consumables Required for<br />

** Use only the PCR plate, adhesive film and thermal cyclers listed<br />

in Table 4.1 on page 40.
