To elute the PCR products:<br />

1. When the wells are completely dry after the third wash, turn off<br />

the vacuum.<br />

2. Carefully remove the plate from the manifold and immediately:<br />

A. Blot the bottom of the plate on a thick stack of clean absorbent<br />

paper to remove any remaining liquid.<br />

B. Dry the bottom of each well with an absorbent wipe.<br />

Immediately blot the bottom of the plate and dry the bottom of<br />

each well. Any remaining liquid will quickly seep back into the<br />

wells.<br />

3. Aliquot 5 mL RB Buffer to a solution basin.<br />

4. Using a 12-channel P200 pipette, add 45 µL RB buffer to each<br />

well of the plate.<br />

5. Tightly seal the plate.<br />

6. Load the plate onto a Jitterbug plate shaker.<br />

7. Set the Jitterbug to setting 5 and moderately shake the plate for<br />

10 minutes at room temperature.<br />

This setting (approximately 1000 rpm) allows as much movement<br />

as possible without losing liquid to the sides of the wells and film.<br />

8. Transfer 45 µL of each eluted sample from the Clontech Clean-Up<br />

Plate to the corresponding well of a fresh 96-well plate following<br />

these guidelines:<br />

• Use a 12-channel P200 pipette set to 60 µL.<br />

• Tilt the Clontech Clean-Up Plate at a 30 to 45 degree angle to<br />

move the liquid to one side of the well.<br />

Optional: use a plate support to keep the plate tilted at an angle<br />

(Well Plate Stand: Diversified Biotech, P/N WPST-1000).<br />

• Pipette up and down 3 to 4 times before removing and<br />

transferring the eluate to a fresh 96-well reaction plate.
