DILUTE THE SAMPLES<br />

To dilute the samples:<br />

8. Place the AccuGENE® Water on ice 20 minutes prior to use.<br />

1. When the <strong>500K</strong> Ligate program is finished, remove the plate and<br />

spin it down at 2000 rpm for 30 sec.<br />

2. Place the plate in a cooling chamber on ice.<br />

3. Dilute each reaction as follows:<br />

<strong>500K</strong> Ligate Program<br />

Temperature Time<br />

16ºC 180 minutes<br />

70ºC 20 minutes<br />

4ºC Hold<br />

A. Pour 10 mL AccuGENE® water into the solution basin.<br />

B. Using a 12-channel P200 pipette, add 75 µL of the water to<br />

each reaction.<br />

The total volume in each well is 100 µL.<br />

Reagent Volume/Sample<br />

Ligated DNA 25 µL<br />

AccuGENE® water 75 µL<br />

Total 100 µL<br />

4. Seal the plate tightly with adhesive film.<br />

5. Vortex the center of the plate at high speed for 3 sec.<br />

6. Spin down the plate at 2000 rpm for 30 sec.<br />

It is crucial to dilute the ligated DNA with AccuGENE® water prior<br />

to PCR.
