REAGENTS REQUIRED<br />

The following reagents are required for this stage. Refer to<br />

Appendix A, Reagents, Equipment, and Consumables Required for 96-Well<br />

Plate Protocol for vendor and part number information. The amounts<br />

listed are sufficient to process one full 96-well reaction plate.<br />

1 vial GeneChip® DNA Labeling Reagent (30 mM)<br />

1 vial Terminal Deoxynucleotidyl Transferase (TdT; 30 U/µL)<br />

2 vials Terminal Deoxynucleotidyl Transferase Buffer<br />

(TdT Buffer; 5X)<br />

IMPORTANT INFORMATION ABOUT THIS STAGE<br />

To help ensure the best results, carefully read the information below<br />

before you begin this stage of the protocol.<br />

• To minimize sample loss due to evaporation, be sure that the plate<br />

is tightly sealed before running the <strong>500K</strong> Label thermal cycler<br />

program.<br />

PREPARE THE REAGENTS, CONSUMABLES AND OTHER COMPONENTS<br />

Thaw Reagents<br />

Thaw the following reagents on ice:<br />

• 5X TdT Buffer<br />

• GeneChip® DNA Labeling Reagent<br />

Leave the TdT enzyme at –20 °C until ready to use.
