90 Jelica Balaž, Aleksandra Smiljanić Artificial inoculations of C. japonica and C. horisontalis young shoots of, immature pear and P. cerasifera fruits and tobacco leaves proved the pathogenicity of investigated strains. The first symptoms on young shoots as dark-brown, elliptical spots 1-2 cm in diameter were observed 4-5 days after inoculations. The spots enlarged and spreaded to base of leaves (Fig.3). Inoculated shoots were necrotic with bended tips after 10-12 days (Fig.4). Dark green to brown spots mainly with bacterial ooze were formed on immature P.cerasifera (Fig. 5) and pear fruits (Fig. 6) 3-4 days after inoculation. All strains caused hypersensitive reaction in tobacco leaves after 24 hr. Fig. 3 – Chaenomeles japonica. Bacterium is rod shaped, gram-negative and asporogenous, facultative anaerob (O/F metabolism). Acid production from glucose was slow in anaerobic but fast in aerobic conditions. Catalase is produced, but not oxidase and arginin dehydrolase. Gelatin liquefaction was positive. Production of H 2 S and indol, reduction of nitrate, digestion of protein (agar plate test), and hydrolysis of starch and urease were negative. Indirect ELISA results showed C. japonica and C. horisontalis strains to be serologically identical to the referent NCPPB 595 and our JK E. amylovora strains, which were used for preparation of polyclonal bacterial antibodies.
