Populations of the non-vector leafhopper, Empoasca kerr, were consistently<br />

higher on Vigna radiata plants infected with mungbean yellow mosaic virus than<br />

on healthy plants. Though total nitrogen was reduced in diseased plants,<br />

different forms of nitrogen were higher. Viral infection also altered amino<br />

acid content, both qualitatively and quantitatively. Diseased leaves had a<br />

lower sugar, magnesium and potassium content, whereas moisture, phosphorus, and<br />

Cytological studies were conducted on yellow mosaic disease in mungbean to<br />

detect the presence of the virus in the host and in the vector Bemisia tabaci<br />

(whitefly). When the epidermal cells from the diseased and healthy leaves were<br />

treated with a fluorescent dye, acridine orange, and examined under fluorescent<br />

microscope, the virus appeared as orange red fluorescing inclusions in the<br />

cytoplasm of the cells from diseased leaf only. Among the various cytological<br />

stains used in light microscopic investigations, Pyronine G-methyl green<br />

combination proved superior over other stains for differentiations between the<br />

nuclei and virus inclusions. The virus was located in the diseased leaf<br />

epidermal cell as red staining masses which were absent in healthy ones. The<br />

accumulation of dark orange staining mass was noticed in the first 2-3<br />

abdominal segments of the whiteflies fed on diseased mungbean plants but not in<br />

the whiteflies fed on healthy plants. [AS]<br />

Differences in total proteins, polyphenol oxidase, and peroxidase between<br />

leaves of mungbean cultivar H 45 (susceptible to mungbean yellow mosaic virus)<br />

and LM 170 (tolerant) were compared. The diseased leaves of both varieties<br />

showed higher activities of polyphenol oxidase and peroxidase. Both starch gel<br />

electrophoresis and DEAE-cellulose column chromatography showed that the<br />

diseased leaves of LM 170 contained a higher number of protein peaks and<br />

isoperoxidases than healthy leaves of the same variety. [AS]<br />
