Themethod required a total of 40 minutes to elute all chlorogenic acidsand re-equilibrate the column (Figure 1). The major componentin the raw material extract is chlorogenic acid and accountsfor almost 50 % of the total content.Kinetex Core-Shell Technology allows scientists to achieve substantiallyhigher chromatographic efficiencies at much lower pressuresthan the equivalent fully porous material. Kinetex core-shellcolumns enhance the performance of any existing HPLC platform,including UHPLC systems.For those labs that have older HPLC systems with pressure limitations,Kinetex 5 µm core-shell columns allow for substantial improvementin chromatography.The increased efficiency of the Kinetex core-shell 5 µm columnallowed us to reduce the column length to 100 mm and achievesimilar if not better resolution. When the gradient was scaled tothe new column dimensions, chlorogenic acid eluted in only 4.9minutes and the total analysis was completed in only 15 min, a64 % reduction in total analysis time. The corresponding decreasein peak width due to the increase in column efficiency caused thechlorogenic acid peak to go beyond the linear range of the UVdetector (Figure 2). Samples had to be diluted in order to bringthe signal intensities within the linear range. When analyzing botanicalsand nutraceuticals, the determination of content in a finishedproduct formulation can be very challenging.
