It is generally used at a workingconcentration of 0.25%, prepared in a balancedsalt solution, without calcium or magnesium, at apH of 7.6-7.8. After treatment, trypsin activitycan be halted by adding a serum media mixture,or a <strong>Trypsin</strong> inhibitor (from soybean, forexample) in serum-free systems.• <strong>Trypsin</strong>-<strong>Versene</strong> (17-161) or <strong>Trypsin</strong>-<strong>EDTA</strong>(ethylenediaminetetraacetic acid) is a combinedreagent used to remove cells from the culturevessel surface into suspension. The <strong>EDTA</strong>, achelator of divalent cations, is added to trypsinsolutions to enhance activity. The calcium andmagnesium in the extracellular matrix, with aidsin cell-cell adhesion, also obscures the peptidebonds that trypsin acts on. The <strong>EDTA</strong> is addedto remove the calcium and magnesium from thecell surface which allows trypsin to hydrolyzespecific peptide bonds. This activity can behalted by adding a serum media mixture, or a<strong>Trypsin</strong> inhibitor (from soybean, for example) inserum-free systems. It is supplied at usedilution.• <strong>Versene</strong> (17-711) or <strong>EDTA</strong> (ethylenediaminetetraaceticacid) is a chelating agent used to liftsome cell types from the culture vessel surfaceinto suspension. It can be used as a wash priorto the addition of a trypsin product or used asdescribed above.
