4 | KST <strong>Re</strong>-<strong>introduction</strong> <strong>Guidelines</strong>BACKGROUNDThe <strong>Kihansi</strong> <strong>Spray</strong> <strong>Toad</strong> (KST),Nectophrynoides asperginis, is endemicto 2.0 hectares of spray zone in the <strong>Kihansi</strong>Gorge of south-central Tanzania. The toadwas believed to be extirpated from the areain 2003 and was declared Extinct in the Wildby the IUCN in October 2009. The species’rapid decline followed dam constructionfrom 1996–2000 and coincided withthe emergence of the amphibian chytridfungus in the population and the flushingof sediments into the gorge in June 2003.Lower <strong>Spray</strong> Wetland, spray irrigation system, and the Mhalala bridgeover the <strong>Kihansi</strong> River. (credit: William Newmark)In November 2000, at the invitation of theTanzanian Government, 499 toads werecollected and transferred to the Bronx Zooto initiate a captive breeding program.The captive population has experiencedperiodic husbandry and health challenges,but currently over 6,000 toads are housedby Bronx and Toledo Zoos.A Population and Habitat ViabilityAssessment (PHVA) workshop held inBagamoyo Tanzania from 14–17 May2007 brought together a diverse rangeof stakeholders and experts to produceguidance on how KST managementchallenges could be handled by way ofdeveloping a KST recovery plan. This PHVAremains highly relevant and forms thebackbone of this re-<strong>introduction</strong> plan. Oneof the primary challenges addressed duringthe 2007 PHVA meeting was increasingthe size of the captive population, whichhas suffered several crashes and was, at thetime, the same size as when it was founded.Since this meeting, the captive populationhas grown, providing a large enough stockmaintained in biosecure facilities to returnapproximately 4,000 animals to Tanzaniaannually.This document is designed to guide there-<strong>introduction</strong> of KST to the <strong>Kihansi</strong>Gorge. The guidelines provided herein area product of the <strong>Kihansi</strong> <strong>Spray</strong> <strong>Toad</strong> <strong>Re</strong><strong>introduction</strong>Workshop that was held in Dares Salaam during three days in February2010. The workshop was facilitated byCuthbert Nahonyo from the Universityof Dar es Salaam and attended by over 76people. Several Tanzanian governmentaland nongovernmental entities wererepresented including the Lower <strong>Kihansi</strong>Environment Management Project(LKEMP) and other programs within theNational Environment Management Council(NEMC), Tanzania Wildlife <strong>Re</strong>searchInstitute (TAWIRI), Division of Wildlife,TANESCO, University of Dar es Salaam,and Sokoine University. Internationalparticipants included representatives fromWCS-Bronx Zoo, Toledo Zoo, San DiegoZoo, <strong>Amphibian</strong> Ark, Museo Tridentino diScienze Naturali, North-West University,IUCN/SSC <strong>Re</strong>-<strong>introduction</strong> SpecialistGroup, IUCN/SSC <strong>Amphibian</strong> SpecialistGroup, State University of New York, USGSPatuxent Wildlife <strong>Re</strong>search Center, and theWorld Bank (see Appendix 1 for a completelist of participants).The first day of the re-<strong>introduction</strong>workshop focused on presentations thatsummarized research findings and statusof conservation work since the PHVAmeeting in 2007. Topics covered includedvegetation changes in the gorge, status ofcaptive populations, contaminant levelsin the gorge, probiotic research underwayin Tanzania, histopathology research in


KST <strong>Re</strong>-<strong>introduction</strong> <strong>Guidelines</strong> | 5Tanzania, review of global amphibiandisease research, hydrology of the gorge,invertebrates in the gorge, and riskmitigation for the shipment of toads to Darbreeding facility. The deliberations of thefirst day of the workshop are presented inVolume I of the proceedings.On the second day, participants split intoworking groups to discuss what work isneeded in the areas of captive populationmanagement in Tanzania, pathologyscreening of KST to minimize risks ofintroducing harmful pathogens into thegorge, experimental re-<strong>introduction</strong>s (akasoft releases) and the longer term initiativeto reestablish a population in the gorge,ecological monitoring of the gorge habitat,and the coordinated administrationwithin and among relevant agencies ofthe Tanzanian government (particularlyNEMC), and social dimensions of theproject (national and international presscommunications, and local educationand engagement with the <strong>Kihansi</strong> areacommunity).Working groups presented summariesof the key needs and actions within theirareas on the third day of the workshop, andopened each topic up for broader reviewand discussion by all participants. Finally,a timeline was presented that organizedthe initiation dates for major actions thatwere determined over the course of theworkshop.


6 | KST <strong>Re</strong>-<strong>introduction</strong> <strong>Guidelines</strong>KIHANSI SPRAY TOADRE-INTRODUCTIONGUIDELINESThe goals of the KST re-<strong>introduction</strong> program are to establish a viable,free-ranging population of the KST within the <strong>Kihansi</strong> Gorge, togetherwith protection and restoration of habitat components critical to the survivalof KST populations in the gorge.The re-<strong>introduction</strong> requires a multidisciplinary approach involving a teamof persons drawn from a variety of backgrounds with appropriate expertise.Team leaders are responsible for coordination between the various bodiesand provision has been made for publicity and public education aboutthe project. Above all, it is recommended that the re-<strong>introduction</strong> beadaptively managed, thereby learning from successes and failures as therelease program proceeds.

No subsequent outbreaks, individual cases,or PCR positive animals since that time.Ranavirus: No histologic findings suggestive ofranaviral disease in captive population.Enteric Parasites: Sporadic presence of entericnematodes (strongyloides-like) in both wild andcaptive populations.<strong>Re</strong>spiratory Parasites: Animals came into theUS with Rhabdias (lungworm) present. No recentnecropsy evidence of parasite (since March 2002).Intravascular Ciliates: Intermittent presence ofintravascular ciliates in TolZ collection. Activity 2KST <strong>Re</strong>-<strong>introduction</strong> <strong>Guidelines</strong> | 9Complete screen of captive US population for pathogens of specific concern for permit process.Output: Screening to be completed by AP, DM, and CH. Summary of results to date and plans foradditional testing below.Chytrid: Extensive screening by taqman PCRnegative (Boyle technique) and no cases documentedon histology other than single outbreak (see ActivityOne). Based on these findings, the captive USpopulation is considered to be free of the Chytridfungus.Ranavirus: Does occur sporadically in USzoos, but neither Bronx nor TolZ have had casesin their amphibian collections, let alone in theKSTs. But with exposure to rest of collection, it isTHEORETICALLY possible the KSTs were exposedat some point or even came from TZ with it.Screening to date: No suspicious lesions havebeen noted in any of over 556 full necropsies (seeActivity One). Targeted surveillance of organscollected at necropsy by PCR have revealed a smallnumber of tests that were questionably positive oninitial tests (2/11 animals at Bronx without DNAsequencing) but were negative on subsequenttesting in a different laboratory. Twenty-five animalsfrom TolZ (healthy animals culled for diseasesurveillance) were recently tested and all negative byTaqman PCR (San Diego Zoo lab).Additional testing planned: Run Taqman PCRon banked (frozen) 40 to 70 randomly selectedsamples from Bronx and possibly some from TolZ.Run Taqman PCR on another diagnostic cull fromTolZ.Enteric parasites: A strongyloides-like nematodeis present in the captive collection, that may havebeen acquired in captivity or been present in thewild population.Additional work planned: DM to reviewhistorical parasitology results from CornellUniversity at the time KST came into captivity. Thiswill help to determine origin of the parasite (captivevs wild). Collect appropriate samples from TolZdiagnostic cull (same as above) to aid in specificidentification of the enteric nematode currentlypresent in the captive population.Intravascular ciliates: This appears to be a freelivingorganism that can cause opportunistic disease.It was first identified in October 2003 and has beensporadically diagnosed via histology in captive KSTs(49 necropsy cases) since that time. Appears to be adensity-dependent problem and has only been seenin a small number of KST in the TolZ population.Additional work planned: CH to review casesin order to assess if organism limited to some tanksor is widespread in collection. <strong>Toad</strong>s may comeback to TZ with the intravascular ciliates present.However, the persistence in KST and the potentialsignificance of this organism in other species areunknown. Will continue to monitor the organism,especially during the challenge experiments.Additional discussion points:JM – Discussion about keeping low levels ofparasites to avoid loss of immunity.AP – May be helpful to maintain a low level ofparasites endemic to the site of release.DM – Only about 30/500 cases show enteritis(reactions to parasitism).Discussion about likelihood of parasitism(nematodiasis) being animal density dependent.Nematodiasis, coccidiosis (seen very rarely incaptive animals), and other parasites may be less ofa concern in the wild due to more space and abilityto move.Diseases and PathologyTime frame: Complete disease screening process for permit by April 30th.Organization structure and information management: <strong>Re</strong>sults to date discussed betweenveterinary advisors of KST program at workshop. Protocols willbe based on existing protocols used in the US KST program (McAloose, Hanley), protocols outlinedin the amphibian disease control manual (Pessier AP & Mendelson JR. 2010. A manual for control ofinfectious diseases in amphibian survival assurance colonies and re<strong>introduction</strong> programs.Version 1.0 Captive Breeding Specialist Group) and research protocols developed by SokoineUniversity of Agriculture (Misinzo G & Malago J 2009. A survey of infectious amphibian diseasesaffecting free-ranging amphibians inhabiting <strong>Kihansi</strong> gorge and University of Dar es Salaamcompound). Additionally, create protocol for sample transfer to SUA.Output: KST veterinary advisory group to assign tasks within group.Time frame: Protocols in place by June 30th.Organization structure and information management: Protocols to be distributed to facilitieshousing captive assurance populations of KST and those doing diagnostic work.Activity 4Cross training of Pathology and Microbiology/Molecular diagnostics experts prior to and inpreparation for return of KSTs to Tanzania.Output: Discussion among veterinary advisors to KST workshop. Discussed purposeof cross training, length of stay, and what to study.For JM, training will be intensive and focusedto enhance existing histopathology knowledgeand develop specialized expertise in amphibianDr. Chris Hanley, Toledo zoo veterinarian, instructing University ofDar es Salaam captive breeding technicians.histopathology with an emphasis on normalhistology and amphibian diseases and grossnecropsy of the KST and other amphibians. Onemonth of training will be split into two-week blockswith two weeks each at the WCS and ZSSD. Initialtraining in basic normal amphibian histology anddiseases will occur at SDZ to be followed by KSTfocused and additional amphibian disease trainingat WCS. Training will include accessing existingdigital catalog of images/baseline and acquiringamphibian pathology related references.For GM, intensive training will focus on enhancingexisting knowledge in chytrid culture andpolymerase chain reaction (PCR). 1 week of trainingat the North-West University in South Africa withChe Weldon in both activities (chytrid culture andconventional PCR) will be the priority; an additional1 week of training at SDZ (real time-PCR) is alsorecommended. Training will include discussion andprovision of positive controls and quality controlprotocols to supplement existing SUA protocols forchytrid culture and PCRThe veterinary advisory group strongly recommendsthat all cross-training be prioritized and occur priorto KST return to TZ.Time frame: Initial cross training to be completed by May 30, 2010. Diagnostic support andcollaboration between US and TZ experts to be ongoing through 7 month and 5 year time frames.Organization structure and information management: Travel arrangements for initial crosstraining to be completed between essential participants for each cross training trip (DM and AP withJM; CW and AP with GM).

Diseases and Pathology12 | KST <strong>Re</strong>-<strong>introduction</strong> <strong>Guidelines</strong>Activity 6Develop and perform experiments that expose amphibian species currently present in the <strong>Kihansi</strong>Gorge to captive-reared KST and/or substrates that have been exposed to captive-reared KST.The KST population has previously been exposed to the cosmopolitan amphibian collections at theUS institutions. This increases the risk that the captive KST may introduce a novel pathogen into the<strong>Kihansi</strong> Gorge. Therefore, before re<strong>introduction</strong> (“into the wild”) can occur, disease transmissionexperiments MUST be undertaken. This should be done early in the process to avoid holding upre<strong>introduction</strong> process if a problem was identified.Because of the need in performing these experiments in a narrow time frame, they may need to becarried out at the biosecure facility at the University of Dar es Salaam. <strong>Re</strong>gardless of location, theseexperiments should be started when the captive TZ KST population is stable.Logistics of experiments: husbandry and care of other gorge species, space requirements, ability tohouse multiple species together, and numbers needed and that can be acquiredOutput: A general outline of the suggested experiments is provided below. The finalexperimental design will be determined once additional information has been gathered.• Experiments should be carried out using multipledifferent amphibian species from the <strong>Kihansi</strong>Gorge if possible. This is because differentspecies may vary in susceptibility to differentpotential pathogens.• It needs to be determined if exposure of Gorgeamphibians will occur by direct housing withcaptive-reared KST (direct exposure) or ifGorge amphibians will be exposed to substratespreviously used in KST enclosures (indirectexposure). Other Gorge amphibians do notusually inhabit spray zones and therefore maynot tolerate husbandry conditions used tomaintain KST. A final decision will be made inconsultation with amphibian husbandry experts.• Experiments will require appropriatecontrols. At a minimum, suggested enclosurearrangements include: 1) Enclosure with Gorgeamphibians not exposed to captive-reared KST orsubstrates exposed to captive KST; 2) Enclosurewith Gorge amphibians exposed to captive-rearedKST or substrates exposed to captive KST. Asnoted in # 1, multiple concurrent experimentsmay be performed using different amphibianspecies from the <strong>Kihansi</strong> Gorge. The number ofanimals used in the exposure experiments willbe determined by availability of gorge amphibianspecies, facility space and by consultation with anepidemiologist.• If substrate experiments are performed:Enclosure(s) of current gorge amphibiansand enclosure(s) of KST are establishedand maintained for 3-4 weeks. At the endof this time period substrates from the KSTenclosure are moved to enclosures containinggorge amphibians. Substrates from the gorgeamphibians could also be introduced to theKST enclosures to evaluate any potential forpathogens present in gorge amphibians to affectreintroduced KST.• Both direct and indirect (substrate exposure)experiments should be conducted for aminimum of 60-90 days or until all animals havedied. Animals still living after the end of theexperimental period will be euthanized.• Necropsies will be performed on all animalsfrom both exposed and control groups that dieduring the course of the experiment or that areeuthanized at the end of the experimental period.Necropsies will include histopathology andcollection of tissues for use in ancillary diagnostictests (e.g. PCR or virus isolation). Comparisonsof mortality, histologic findings and ancillarydiagnostic findings between control and exposedgroups of animals will be performed to assess riskof introducing a novel pathogen to the <strong>Kihansi</strong>gorge.• Consideration may be given to performingexperiments that place gorge amphibiansunder “stress” (e.g. overcrowding; suboptimaltemperature) in order to maximize the possibilitythat infectious diseases will be expressed.Consider experiment repeated at <strong>Kihansi</strong> facilityusing gorge water and native substrate (or transfernative substates from <strong>Kihansi</strong> Gorge to the Darfacility) to assess ability of KST to survive afterrelease.Timeframe: Ideally, experiment can be started within 2-3 months after assurance populationstabilizes IF experiments are done at Dar facility. If timeline for re<strong>introduction</strong> changes, then mayhave to run before population stabilizes, as these experiments MUST be done before re<strong>introduction</strong>s.Organization structure and information management: North-West University (C.
