ON SRD 1p36 / SE 1(1qh)Neuroblastomas frequently have deletions of chromosome1p and amplification of the N-myc oncogene. These deletionstend to be large and extend to the telomere, but a commonregion within sub-band 1p36.3 is consistently lost. Inactivationof a tumor suppressor gene within 1p36.3 is believed to beassociated with an increased risk for disease relapse. The1p36 specific DNA probe has recently been changed to coverthe recently described smallest region of consistent deletion(SRD) between D1S2795 and D1S253.The SRD (1p36) specific DNA probe is optimized to detectcopy numbers of the 1p36 region on chromosome 1. Thechromosome 1 satellite enumeration probe (SE 1) at 1qh isincluded to facilitate chromosome identification.ON MLL (11q23) / SE 11Deletions of the long arm of chromosome 11 (11q) havebeen noted in primary neuroblastomas. It is assumedthat a tumor suppressor gene mapping within 11q23.3 iscommonly inactivated during the malignant evolution ofa large subset of neuroblastomas, especially those withunamplified MYCN.The MLL (11q23) specific DNA probe is optimized to detectamplification or deletion involving the MLL gene region at11q23 in a dual-color assay on metaphase/interphase spreads,blood smears and bone marrow cells.
