Proceed<strong>in</strong>g of the 4 rd International Symposium“NEW RESEARCH IN BIOTECHNOLOGY” USAMV Bucharest, Romania, 2011If Monascus purpureus was <strong>de</strong>veloped on gra<strong>in</strong> milk, the content of citr<strong>in</strong><strong>in</strong> <strong>in</strong>culture media which result after fermentation is 386 mg/L [4-5]. If the same stra<strong>in</strong> wasgrowth on potato-glucose medium, the citr<strong>in</strong><strong>in</strong> content at the end of fermentation is lessthan 1.1 ppm.If we reffer<strong>in</strong>g to raw materials used <strong>in</strong> food <strong>in</strong>dustry, maximum concentration ofcitr<strong>in</strong><strong>in</strong> f<strong>in</strong>d <strong>in</strong> cereals was 224.6 ng/g.In some products used <strong>in</strong> animal fod<strong>de</strong>r, sometimes citr<strong>in</strong><strong>in</strong> concentration is bigger that 405ppb [5].In the case of Monascus sp.1 which <strong>de</strong>velop <strong>in</strong> culture media with <strong>de</strong>xtrose, thestudy regard<strong>in</strong>g the content of citr<strong>in</strong><strong>in</strong> <strong>in</strong> liquid culture media <strong>in</strong>dicate a maximumconcentration at 96 h of biosynthesis, when the content of citr<strong>in</strong><strong>in</strong> from the system atta<strong>in</strong>7.72 ppm. After 120 h of biosynthesis, the content of citr<strong>in</strong><strong>in</strong> from the system <strong>de</strong>creased to3.5 ppm (figure 17) and cont<strong>in</strong>ue to <strong>de</strong>crease until 2,68 ppm at 240 h of biosynthesis.This fact is positive because the maximum concentration of red azaphilones wasatta<strong>in</strong>ed after 200h, when <strong>in</strong> the system the citr<strong>in</strong><strong>in</strong> concentration is less than 2.7 ppm. Inthis case results obta<strong>in</strong>ed are comparable with citr<strong>in</strong><strong>in</strong> content from juice or w<strong>in</strong>e.In the case of stra<strong>in</strong> Monascus sp. 2, the analysis <strong>in</strong>dicate a maximumconcentration of citr<strong>in</strong><strong>in</strong> of 24.48 ppm, at 120h of biosynthesis. After 168h the content ofcitr<strong>in</strong><strong>in</strong> from the system <strong>de</strong>creased at 3.39 ppm figure 18). After 10 day of fermentation, thecontent of citr<strong>in</strong><strong>in</strong> from the system <strong>de</strong>creased to 2.2 ppm.If we compare the time at which we obta<strong>in</strong> the maximumum content of azaphylonewith time <strong>in</strong> which we obta<strong>in</strong> the m<strong>in</strong>imum content of mycotox<strong>in</strong>, we obta<strong>in</strong> an optimaltime of biosynthesis situated <strong>in</strong> the range of 200-220h.4. CONCLUSIONGood results are obta<strong>in</strong>ed <strong>in</strong> the case of biosynthesis of azaphilone on <strong>de</strong>xtrose culturemedia us<strong>in</strong>g Monascus sp.1 or Monascus sp.2 as stra<strong>in</strong>s. In the last case, the maximumconcentration of red azaphilones are obta<strong>in</strong>ed after 170 h of biosynthesis, (OD =1 for redazaphilones) when the content of citr<strong>in</strong><strong>in</strong> is situated un<strong>de</strong>r 3 ppm. These results recommendthe stra<strong>in</strong>s Monascus sp.1 and Monascus sp.2 to obta<strong>in</strong> food additives (pigments) <strong>in</strong>submerged culture media with <strong>de</strong>xtrose.ACKNOWLEDGEMENTS.
