Proceed<strong>in</strong>g of the 4 rd International Symposium“NEW RESEARCH IN BIOTECHNOLOGY” USAMV Bucharest, Romania, 2011(STD). The <strong>de</strong>tection limit, DL was estimated by multiply<strong>in</strong>g the STD by a factor of 3, andthe quantification limit, QL was <strong>de</strong>f<strong>in</strong>ed as 10 times the STD. The values obta<strong>in</strong>ed were0.008 mg/L for DL and 0.026 mg/L for QL.The precision or the <strong>de</strong>gree of reproducibility is expressed as the coefficient ofvariation (CV). Precision was <strong>de</strong>term<strong>in</strong>ed by perform<strong>in</strong>g six replicate analyses of the samesample on six different days. The value obta<strong>in</strong>ed for precision was 9.8 % for direct <strong>in</strong>jectionof the sample <strong>in</strong> chromatographic system.The uncerta<strong>in</strong>ty was estimate accord<strong>in</strong>g EURACHEM/CITAC. This estimationaccounts for the most significant sources of uncerta<strong>in</strong>ty that are thought to <strong>in</strong>terfere <strong>in</strong> thef<strong>in</strong>al result. In this work, the contributions to uncerta<strong>in</strong>ty were ascribed to: the uncerta<strong>in</strong>ty<strong>in</strong> the preparation of standards, the uncerta<strong>in</strong>ty of the calibration curve and the uncerta<strong>in</strong>ty<strong>in</strong> the precision of the method. The global uncerta<strong>in</strong>ty was estimated at 23.5 % forconcentration above 0.25 mg/L.Quantification of trans-resveratrol <strong>in</strong> Romanian w<strong>in</strong>eThe <strong>de</strong>term<strong>in</strong>ation of the trans-resveratrol alone proved to be accurate us<strong>in</strong>g asimple procedure, without sample pre-treatment other than filtration, allow<strong>in</strong>g for a lessthan 15 m<strong>in</strong>ute analysis run time.However, the chromatograms of the standards are free from <strong>in</strong>terferences, with thepeak of trans-resveratrol clearly separated (Fig. 3a). Regard<strong>in</strong>g the w<strong>in</strong>e samples it isnoticeable that <strong>in</strong> the red w<strong>in</strong>e (Fig. 3b) the <strong>in</strong>terferences are more significant than <strong>in</strong> whitew<strong>in</strong>es (Fig. 3c), <strong>in</strong> spite of the lower amounts found <strong>in</strong> the latter samples.In the work of Ratola et all [9], Portuguese red w<strong>in</strong>e from different regions hadtrans-resveratrol content from 0.55 to 1.24 mg/L and white w<strong>in</strong>es had values rang<strong>in</strong>g fromnot <strong>de</strong>tected to 0.19 mg/L. Other studies report trans-resveratrol content rang<strong>in</strong>g from 0.508to 2.534 mg/L for greek w<strong>in</strong>es [10] and values ranged from 0.25 to 8.87 mg/L for Italianw<strong>in</strong>es [11].The amount of trans-resveratrol <strong>in</strong> the different analyzed w<strong>in</strong>es variedconsi<strong>de</strong>rably <strong>de</strong>pend<strong>in</strong>g on the grape variety, environmental factors <strong>in</strong> the v<strong>in</strong>eyard andw<strong>in</strong>e-process<strong>in</strong>g techniques. In our study, the occurrence of trans-resveratrol was confirmed<strong>in</strong> all of the analyzed 14 red w<strong>in</strong>e samples, with concentration rang<strong>in</strong>g between 0.025 and6.598 mg/L (Table 1). For the white w<strong>in</strong>e, 5 of the 10 tested revealed the presence of thecompound at concentration between 0.132- 0.159 mg/L (Table 2).It is known that trans-resveratrol is produced by grape berries <strong>in</strong> response to fungal<strong>in</strong>fection and UV irradiation [12]. Thus, it is possible that sun exposure of grapes grown atMuntenia region may be a factor <strong>in</strong> <strong>in</strong>creased resveratrol level compared with the grapesfrom Oltenia region. In addition, the red grape varieties Feteasca Neagra may be geneticallyricher <strong>in</strong> this compound than other varieties.Graphical representation of mean concentration of trans-resveratrol found <strong>in</strong> thisresearch, <strong>de</strong>pend<strong>in</strong>g on the red w<strong>in</strong>e variety show that the content of trans-resveratrol<strong>de</strong>creased <strong>in</strong> or<strong>de</strong>r Feteasca Neagra, Merlot, P<strong>in</strong>ot Noire, Cabernet Sauvignon (Fig. 4).Concern<strong>in</strong>g white w<strong>in</strong>e, higher concentration of trans-resveratrol was found <strong>in</strong> MuscatOttonel variety.179
