times <strong>and</strong> tested for coupling activity towards a-chymotrypsinogen (A) <strong>and</strong> glycyl-leucine (B).<br />

Coupling at low pH is less efficient, but may be advantageous if the lig<strong>and</strong> loses biological<br />

activity when it is fixed too firmly, e.g. by multi-point attachment, or because <strong>of</strong> steric<br />

hindrance between binding sites which occurs when a large amount <strong>of</strong> high molecular<br />

weight lig<strong>and</strong> is coupled. Use a buffer <strong>of</strong> approximately pH 6.<br />

IgG is <strong>of</strong>ten coupled at a slightly higher pH, for example in 0.2–0.25 M NaHCO 3 ,<br />

0.5 M NaCl, pH 8.5–9.0.<br />

<strong>and</strong> sanitization procedures. Stability data refers to the coupled medium provided that the lig<strong>and</strong> can withst<strong>and</strong> the pH.<br />

Storage<br />

Store the freeze-dried powder below +8 °C in dry conditions.<br />

Store the column in a solution that maintains the stability <strong>of</strong> the lig<strong>and</strong> <strong>and</strong> contains a<br />

bacteriostatic agent, for example, PBS, 0.05% NaN 3 , pH 7.2 or 20% ethanol in a suitable<br />

buffer.<br />

The pH stability <strong>of</strong> the medium when coupled to the chosen lig<strong>and</strong> will depend upon the<br />

stability <strong>of</strong> the lig<strong>and</strong> itself.<br />
