HHMI video, “Polymerase Chain Reaction,” and/or the <strong>DNA</strong>i.org interactive<br />

tu<strong>to</strong>rial on PCR. See Computer Equipment, Files, Software, and Media in the<br />

Materials section above for URLs.<br />

Procedure<br />

9. Show Slide #6, “Lab 2: Copying the <strong>DNA</strong> <strong>Barcoding</strong> Gene Using Polymerase<br />

Chain Reaction (PCR)” as students enter the room.<br />

10. Show Slide #7, and remind students that their overall goal is <strong>to</strong> obtain <strong>DNA</strong><br />

sequence data from the species they are studying. In the last experiment,<br />

students purified <strong>DNA</strong> from their samples. Today, they will amplify, or copy,<br />

the <strong>DNA</strong> barcoding gene using polymerase chain reaction (PCR).<br />

Amplify: In PCR, <strong>to</strong> amplify is <strong>to</strong> increase<br />

in copy number.<br />

11. If students are not already familiar with PCR, it is strongly suggested that<br />

you show the online video, “Polymerase Chain Reaction,” freely available<br />

from Howard Hughes Medical Institute (HHMI). The video is 1 minute 27<br />

seconds long and requires an internet connection and speakers. It is available<br />
