Anti-inflammatory activity<br />
Numerous investigati<strong>on</strong>s have assessed the anti-inflammatory effects of<br />
the c<strong>ru</strong>de d<strong>ru</strong>g in vitro and in vivo (20–22). In <strong>on</strong>e study, a suspensi<strong>on</strong> of<br />
powdered root (1 g/kg suspended in 2% acacia gum, 50 mg/ml) administered<br />
intragastrically to rats for 3 days, 1 hour before the injecti<strong>on</strong> of<br />
Freund’s complete adjuvant, reduced inflammati<strong>on</strong>. Many se<strong>ru</strong>m proteins<br />
such as 2-glycoprotein, a major acute inflammatory phase protein and<br />
pre-albumin were decreased, indicating a reducti<strong>on</strong> in acute inflammati<strong>on</strong><br />
(20). In another study by the same research group, 1 g/kg of the root suspensi<strong>on</strong><br />
reduced the 2-macroglobulin in the se<strong>ru</strong>m of rats given subplantar<br />
injecti<strong>on</strong> of carrageenan suspensi<strong>on</strong> (21).<br />
In <strong>on</strong>e study, air pouch granuloma was induced by subcutaneous injecti<strong>on</strong>s<br />
of 4 ml of carrageenan <strong>on</strong> the dorsum of rats which had been<br />
subcutaneously injected 1 day previously with 6 ml of air <strong>on</strong> the dorsum<br />
(22). The powdered c<strong>ru</strong>de d<strong>ru</strong>g was administered by gastric lavage at a<br />
dose of 1 g/kg bw for 3 days. Radioactive sodium sulfate was injected<br />
intraperit<strong>on</strong>eally <strong>on</strong> day 9 and the incorporati<strong>on</strong> of radio-labelled sulfur<br />
in glycosaminoglycan, oxidative phosphorylati<strong>on</strong> (ADP/O ratio), mg 2+ -<br />
dependent-ATPase enzyme activity and succinate dehydrogenase activity<br />
were determined in the mitoch<strong>on</strong>dria of the granuloma tissue. Administrati<strong>on</strong><br />
of the root decreased the glycosaminoglycan c<strong>on</strong>tent of the granuloma<br />
tissue by 92%, compared with 43.6% following treatment with hydrocortis<strong>on</strong>e<br />
(15.0 mg/kg bw) and had no effect following treatment with<br />
phenylbutaz<strong>on</strong>e (100.0 mg/kg bw) (22).<br />
In a further study, the effect of oral administrati<strong>on</strong> of the c<strong>ru</strong>de d<strong>ru</strong>g<br />
(root powder, 1 g/kg bw, daily for 15 days) <strong>on</strong> paw swelling and b<strong>on</strong>y<br />
degenerative changes in Freund’s complete adjuvant-induced arthritis in<br />
rats was assessed. Intragastric administrati<strong>on</strong> of the powdered root to the<br />
rats caused a significant reducti<strong>on</strong> in both paw swelling and degenerative<br />
changes as observed by radiological examinati<strong>on</strong>. The reducti<strong>on</strong>s were<br />
better than those produced by the reference d<strong>ru</strong>g, hydrocortis<strong>on</strong>e<br />
(15.0 mg/kg bw) (16, 23).<br />
Daily intragastric administrati<strong>on</strong> of 1.0 g/kg bw of the powdered root<br />
suspended in 2% gum acacia, was given 1 hour before the inducti<strong>on</strong> of<br />
inflammati<strong>on</strong> by injecti<strong>on</strong> of Freund’s complete adjuvant in rats for<br />
3 days. Phenylbutaz<strong>on</strong>e (100.0 mg/kg bw) was administered to another<br />
group of animals as a positive c<strong>on</strong>trol. Assessment of acute phase reactants<br />
of the blood showed changes in the c<strong>on</strong>centrati<strong>on</strong> of 2-glycoprotein,<br />
major acute phase 1-protein, and pre-albumin in rats with inflammati<strong>on</strong>,<br />
while in animals treated with the powdered root, the 2-<br />
