Radix Withaniae<br />

As a general t<strong>on</strong>ic to increase energy, improve overall health and prevent<br />

disease in athletes and the elderly (1, 16).<br />

Treatment of br<strong>on</strong>chitis, dyspepsia, impotency, scabies and ulcers (5, 16).<br />

Antistress activity<br />

The activity of a standardized extract of the root (1:1 aqueous ethanol<br />

fracti<strong>on</strong> c<strong>on</strong>taining the withanolide glycosides and withaferin A at a<br />

c<strong>on</strong>centrati<strong>on</strong> of 28–30%) was investigated in a rat model of chr<strong>on</strong>ic<br />

stress. A mild, unpredictable foot-shock was administered <strong>on</strong>ce daily for<br />

21 days to rats. These chr<strong>on</strong>ic stress-induced perturbati<strong>on</strong>s were attenuated<br />

by the intragastric administrati<strong>on</strong> of the c<strong>ru</strong>de d<strong>ru</strong>g, at a dose of<br />

25.0 and 50.0 mg/kg body weight (bw) given 1 h before foot-shock for<br />

21 days (17).<br />

In a mouse model of chr<strong>on</strong>ic stress, the antioxidant effects of the root<br />

were assessed in the forced swimming test. Biochemical analysis revealed<br />

that chr<strong>on</strong>ic swimming significantly increased lipid peroxidati<strong>on</strong> and decreased<br />

glutathi<strong>on</strong>e levels in the brains of mice. The animal models also<br />

showed decreased levels of antioxidant defence enzymes, superoxide<br />

dismutase and catalase. Intragastric treatment with an extract of the c<strong>ru</strong>de<br />

d<strong>ru</strong>g, at a dose of 100.0 mg/kg bw significantly reduced lipid peroxidati<strong>on</strong><br />

and restored the glutathi<strong>on</strong>e levels decreased by chr<strong>on</strong>ic swimming in<br />

mice. Further, the treatment increased levels of superoxide dismutase in<br />

the forebrain and increased levels of catalase (18).<br />

A withanolide-free aqueous fracti<strong>on</strong> from the root (13 kg plant material<br />

in 70% ethanol, aqueous fracti<strong>on</strong>) was evaluated for putative antistress<br />

activity. Intragastric administrati<strong>on</strong> of the preparati<strong>on</strong> to immunocompromised<br />

mice for 7 days increased antibody producti<strong>on</strong> with a<br />

median effective dose of 40 mg/kg bw. Thus, the fracti<strong>on</strong> exhibited significant<br />

antistress activity in a dose-related manner. The same fracti<strong>on</strong><br />

was protective against chemically and physically induced stress in rats<br />

and mice (19).<br />

Intragastric administrati<strong>on</strong> of sitoindosides IX and X, at a dose range<br />

of 50–200.0 mg/kg bw also produced antistress activity in albino mice and<br />

rats, and augmented learning acquisiti<strong>on</strong> and memory retenti<strong>on</strong> in both<br />

young and old rats (16).<br />
