Pharmacopeia) was investigated. Additi<strong>on</strong> of 0.1 ml of an ethanol extract<br />

of the bark to a 100-ml bath chamber c<strong>on</strong>taining isolated guinea-pig uterine<br />

strips, reduced normal and resting t<strong>on</strong>e, and decreased the rate and<br />

amplitude of sp<strong>on</strong>taneous c<strong>on</strong>tracti<strong>on</strong>s (15). The antispasmodic effects<br />

were observed 5–15 minutes after additi<strong>on</strong> and when the extract was removed<br />

the c<strong>on</strong>tracti<strong>on</strong>s resumed. A synergistic effect was observed in rat<br />

uteri treated with the fluidextract of the bark and 1 mg papaverine hydrochloride<br />

(16). In a similar test, the essential oil of the bark reduced uterine<br />

c<strong>on</strong>tracti<strong>on</strong>s in isolated rat uteri (c<strong>on</strong>centrati<strong>on</strong> not stated) (17).<br />

An aqueous, 95% ethanol, methanol or fluidextract of the bark reduced<br />

sp<strong>on</strong>taneous uterine c<strong>on</strong>tracti<strong>on</strong>s in the isolated ute<strong>ru</strong>s from female<br />

rats, at a c<strong>on</strong>centrati<strong>on</strong> of 1:200, or 1mM (11, 18, 19). Methanol extracts<br />

of the bark had antispasmodic effects in estr<strong>on</strong>e-primed barium<br />

chloride-stimulated rat uterine horns in vitro (11). Scopoletin, isolated<br />

from the bark had antispasmodic properties in vitro in estr<strong>on</strong>e-primed<br />

and barium chloride-induced rat uteri. The c<strong>on</strong>centrati<strong>on</strong> needed to produce<br />

a 50% decrease in the c<strong>on</strong>tracti<strong>on</strong> amplitude of a single uterine horn<br />

was 0.09 mg/ml. Similar effects were observed in rodents in a study of<br />

oxytocin- and erg<strong>on</strong>ovine-induced uterine c<strong>on</strong>tracti<strong>on</strong>s (20). A fluidextract<br />

of the bark (c<strong>on</strong>centrati<strong>on</strong> not stated) reduced sp<strong>on</strong>taneous c<strong>on</strong>tracti<strong>on</strong>s<br />

in rat uteri in vitro. Additi<strong>on</strong> of a 95% ethanol extract (c<strong>on</strong>centrati<strong>on</strong><br />

not stated) of the bark to the bath medium prevented barium chloride<br />

and acetylcholine-induced spasms in isolated guinea-pig ileum (21).<br />

A methanol extract (100 g dried bark), as well as the ethyl acetate and<br />

butanol fracti<strong>on</strong>s of the bark, inhibited sp<strong>on</strong>taneous c<strong>on</strong>tracti<strong>on</strong>s of isolated<br />

rat uteri by 50% at c<strong>on</strong>centrati<strong>on</strong>s of 2.0–200.0 μg/ml. A decrease in<br />

the t<strong>on</strong>e and amplitude of acetylcholine-induced c<strong>on</strong>tracti<strong>on</strong>s was also<br />

observed. The median inhibitory c<strong>on</strong>centrati<strong>on</strong> was 60 μg/ml for the<br />

c<strong>ru</strong>de extract, 36.0 μg/ml for the ethyl acetate fracti<strong>on</strong> and 50 μg/ml for<br />

the butanol fracti<strong>on</strong>. 2´-p-Coumaroyldihydropenstemide was the active<br />

c<strong>on</strong>stituent (22).<br />
