Die Institut Virion\Serion GmbH wurde 1978 in Würzburg gegründet und hat sich von Beginn an auf die 

Herstellung von in-vitro Diagnostika für die Infektionsserologie spezialisiert. Durch das stetige Wachstum 

musste der Standort bereits zweimal im Laufe der Firmengeschichte verlagert werden, jedoch ist das traditionsbewusste 

Familienunternehmen mit seinen mittlerweile 125 Mitarbeitern der Stadt Würzburg treu geblieben. 

Die Produktpalette umfasst mehrere Nachweissysteme für virale, bakterielle, parasitäre oder mykologische 

Krankheitserreger. Über 50 Antigene werden für die Komplementbindungsreaktion (KBR) zum indirekten 

Erregernachweis bereitgestellt. Mit Hilfe der SERION ELISA classic können die unterschiedlichen Antikörperklassen 

zur Bestimmung des Immunstatus erfasst werden. Im Gegensatz dazu ermöglichen SERION ELISA 

antigen 

den direkten Erregernachweis. 

Immunomat und Gemini dienen zur automatisierten Prozessierung 

von SERION ELISA classic und SERION ELISA antigen 

Immunoassays. 

Sämtliche Komponenten und Endprodukte der Institut Virion\Serion GmbH werden nach nationalen und 

internationalen Qualitätsstandards in Übereinstimmung mit der EG Richtlinie 98/79/EG für in-vitro Diagnostika 

und gemäß der ISO-Norm EN ISO 13485:2003 hergestellt. In Deutschland werden die Produkte 

durch einen eigenen Außendienst vertrieben. Die weltweite Vermarktung in inzwischen über 80 Länder 

wird durch die 1986 gegründete Serion Immundiagnostica GmbH und ein umfassendes Netzwerk von 

Distributoren sichergestellt. Für die effiziente Belieferung des asiatischen und brasilianischen Marktes mit den 

ausschließlich in Würzburg/Deutschland hergestellten Produkten wurden in den Jahren 2003 und 2010 eigene 

Niederlassungen in Peking und Curitiba gegründet.

Progress by Experience and Innovation 

1978 saw the founding of Institut Virion\Serion GmbH in the city of Würzburg in southern Germany. From 

the outset the company has specialized in the manufacture of in vitro diagnostic products for the infectious 

disease serology and has, due to continual growth, twice expanded into new premises over the past 30 years. 

This family run business with its roots in Würzburg has remained in the region and currently provides employment 

and career opportunities for around 125 employees. Our product range offers a selection of analysis methodologies 

to aid the diagnosis of viral, bacterial, parasitic and fungal diseases. The complement fixation 

test (CFT) range alone offers in excess of 50 antigens plus associated reagents and controls for the detection 

of pathogen specific antibodies. SERION ELISA classic products allow for the differentiation of antibody 

classes for disease state and immune status determinations. In contrast, SERION ELISA antigen products 

allow for direct pathogen detection. Immunomat TM and Gemini are capable of automatically processing 

SERION ELISA classic and SERION ELISA antigen immunoassays. 

All products are manufactured exclusively in Würzburg, Germany, by Institut Virion\Serion GmbH in compliance 

with national and international quality assurance standards related to in vitro medical devices such as the 

European Directive IVDD 98/79/EC. The company operates under a quality management system based on the 

latest updated version of ISO 13485:2003. Customers in Germany are served by our sales team of product 

specialists while worldwide sales, which at present encompass over 80 countries, are coordinated through 

our subsidiary Serion Immunodiagnostica GmbH and an extensive network of local distributors. In order 

to provide an efficient service for our products in the Asiatic and Brazilian market we established branch offices 

in Beijing and Curitiba in 2003 and 2010. 

The company’s commitment to innovation combined with a wealth of experience and the willingness to break 

new ground ensures that we are always going forward. One of our most recent product developments is the 

SERION Multianalyt technology. Based on the principles of flow cytrometry, this technology allows for 

the simultaneous testing of multiple parameters while using very small sample quantities. This new methodology 

combines a diagnostic capability of the highest quality with time saving and cost effective solutions for the 

modern laboratory requirements.

Immunoassays for the diagnosis of infectious diseases 

Serologische Untersuchungen leisten einen wichtigen Beitrag zur Diagnose von Infektionserkrankungen. Zu 

diesem Zweck werden Antikörper nachgewiesen, die von den Zellen des Immunsystems als Antwort auf die 

Gegenwart einer fremden Substanz, dem Antigen, gebildet werden und zur Abwehr des Krankheitserregers 

dienen. 

Serological investigations are important in the diagnosis of infectious diseases. The immune system produces 

antibodies as a vital component of the response to infection and the presence of foreign bodies or antigens. The 

detection of these antibodies is utilised in the diagnostic process. 


SERION ELISA classic sind qualitative und quantitative Immunoassays für den Nachweis von humanen Antikörpern 

in Serum, Plasma und ggf. Liquor cerebrospinalis gegen spezifische Antigene von Bakterien, Viren, Pilzen oder 

Parasiten zur Unterstützung bei der Diagnose von Infektionserkrankungen. 

SERION ELISA classic tests are qualitative and quantitative immunoassays for the detection of human antibodies 

directed against specific antigens of bacteria, viruses, fungi and parasites in serum, plasma or, when indicated, 

cerebrospinal fluid, for the diagnosis of infectious diseases. 

Testprinzip 

Der ELISA (Enzyme Linked Immunosorbent Assay) ist ein immunologisches Nachweisverfahren, das sich 

insbesondere in der Infektionsserologie zur Erfassung von Antikörpern bewährt hat. Die Reaktion basiert auf 

der spezifischen Interaktion von Antikörpern mit der nachzuweisenden Substanz. Zu diesem Zweck werden die 

Teststreifen der Miktotiterplatten der SERION ELISA classic mit spezifischen Antigenen von Infektionserregern 

beschichtet, an die in der Patientenprobe vorhandene Antikörper binden. Ein weiterer Antikörper, der zuvor 

mit dem Enzym Alkalische Phosphatase markiert wird, detektiert den so gebildeten Immunkomplex. Dieses 

Enzym katalysiert eine Reaktion, in deren Verlauf das farblose Substrat p-Nitrophenoylphosphat in das farbige 

Produkt p-Nitrophenol umgewandelt wird. Die Signalstärke des Reaktionsprodukts ist proportional zur Menge 

der nachzuweisenden Substanz in der Probe und wird photometrisch erfasst.

Test Principle 

The ELISA (Enzyme Linked Immunosorbent Assay) is an immunoassay, which is particularily suited to the 

determination of antibodies in the field of infectious serology. The reaction is based on the specific interaction 

of antibodies with their corresponding antigen. Therefore, the test strips of the SERION ELISA classic microtitre 

plate are coated with specific antigens of the pathogen of interest. If antibodies in the patient´s serum sample 

are present, they bind to the fixed antigen. A secondary antibody, which has been conjugated with the enzyme 

alkaline phosphatase, detects the immune complex. The colourless substrate p-nitrophenylphosphate is then 

converted into the coloured product p-nitrophenol. The signal intensity of the reaction product is proportional to 

the concentration of the analyte in the sample and is measured photometrically.

Infektionen der Atemwege gehören weltweit zu den häufigsten Erkrankungen, die vor allem durch Viren und 

Bakterien hervorgerufen werden. Der schnelle Nachweis des Erregers ist oft entscheidend für eine effiziente 

Therapie. Die Bestimmung spezifischer Antikörper gegen eine Vielzahl von Erregern respiratorischer Erkrankungen 

mit SERION ELISA classic Immunoassays dient sowohl zur sensitiven Erfassung akuter Infektionen der Atemwege

Respiratory tract infections, particularly those of bacterial and viral aetiology, are among the most common 

diseases worldwide and the timely identification of the causative agent is frequently decisive in determining 

the appropriate therapy. The detection of specific antibodies against a wide range of pathogens responsible 

for respiratory diseases using SERION ELISA classic immunoassays serves as a sensitive identifier of acute 

infections as well as aiding in the differential diagnosis in cases of atypical pneumonia.

Die Aktivität von IgG Antikörpern wird häufig mit dem Immunschutz korreliert. Aus der IgG Aktivität gegen Tetanus

Während der Perinatalphase erworbene Infektionen können schwerwiegende Erkrankungen von Neugeborenen 

zur Folge haben. Aus diesem Grund ist der Nachweis von IgM Antikörpern gegen verschiedene Infektionserreger 

im Rahmen eines Screenings sinnvoll, um infizierte Neugeborene schnell zu identifizieren und rechtzeitig 

therapeutische Maßnahmen einleiten zu können. Zu diesem Zweck wurden die folgenden SERION ELISA classic

Infections acquired in utero may have severe consequences for the fetus and newborn child. Consequently, it is 

advisable, within the framework of a screening program, to test for specific IgM antibodies to certain infectious 

agents in order to rapidly identify infected neonates and facilitate prompt and appropriate medical interventions.

Die von Chlamydien hervorgerufenen Erkrankungen betreffen insbesondere die Schleimhäute im Augen-, 

Atemwegs- und Genitalbereich und können schwerwiegende Schäden zur Folge haben. Unbehandelte 

Chlamydiosen führen nicht selten zu Erblindung, Unfruchtbarkeit oder Arthritis. Infektionen mit C. trachomatis 

sind eine der häufigsten, sexuell übertragbaren Erkrankungen mit bakterieller Ursache. C. pneumoniae kann 

chronischen Husten oder Lungenentzündungen auslösen. C. psittaci ist der Erreger der Ornithose. 

Diseases caused by Chlamydia ssp. affect, in particular, the mucous membranes in the eyes, respiratory tract 

and genital region and can have severe consequences. Untreated Chlamydia infections frequently lead to 

blindness, infertility and arthritis. C. trachomatis infection is one of the most common sexually transmitted 

bacterial diseases, while C. pneumoniae can cause chronic bronchitis or pneumonia and C. psittaci is the cause 

of ornithosis (Psittacosis).

A range of SERION ELISA classic immunoassays are validated for the detection of intrathecally synthesized 

antibodies and so enable the detection and differentiation of inflammatory processes within the central nervous 

system (CNS). To this end the following SERION ELISA classic immunoassays may be used to determine the 

antibody activities against certain infectious agents in paired CSF/serum samples in order to determine the 

CSF/Serum quotient according to the scheme of Prof. Hansotto Reiber and calculate the resulting antibody index

Reagenz zur Entfernung von IgM Rheumafaktoren 

Reagent for the removal of IgM rheumatoid factors 

Rheumafaktoren sind vorwiegend gegen IgG gerichtete IgM Antikörper, die in bis zu 5 % der Serumproben 

gesunder Individuen nachgewiesen werden können. Diese IgM Rheumafaktoren binden bevorzugt an IgG 

Immunkomplexe, so dass bei Anwendung erregerspezifischer IgM Nachweise falsch-positive Ergebnisse 

auftreten können. In Proben mit hohem, erreger-spezifischem IgG Anteil besteht außerdem die Möglichkeit, 

dass bindungsschwächere IgM Antikörper durch bindungsstärkere IgG Antikörper verdrängt werden. Ein IgM 

Nachweis ohne Rf-Absorption kann in solchen Fällen zu falsch-negativen Resultaten führen. Aus diesem Grund 

ist es erforderlich, die Serumproben für die IgM Bestimmung mit Rf-Absorbens vorzubehandeln, welches bis 

zu 15 mg IgG/ml unverdünntes Serum und somit auch die an IgG Antikörper gebundenen IgM Rheumafaktoren 

präzipitiert. 

Rheumatoid factors are predominantly against IgG directed IgM antibodies that can be detected in serum 

samples of approximately 5 % of healthy individuals. Those IgM rheumatoid factors preferably bind to IgG immune 

complexes. Therefore, false positive results may appear in pathogen-specific IgM detection. In samples with a 

high concentration of pathogen-specific IgG antibodies, weakly binding pathogen-specific IgM antibodies may 

be displaced by stronger binding IgG antibodies. In those cases, IgM detection can show false negative results. 

Therefore, it is necessary to pretreat serum samples with rheumatoid factor-absorbent. Prior to pathogenspecific 

IgM detection, it precipitates up to 15 mg IgG/ml undiluted serum and thereby also IgG binding IgM 

rheumatoid factors.

threshold, e. g. in order to exclude high seroprevalences. Therefore, the evaluation of individual SERION 

ELISA controls might deliver borderline results in exceptional cases.

The SERION Multianalyt TM technology allows the simultaneous

Testprinzip 

Die Analysereaktion erfolgt in den Kavitäten einer Mikrotiterplatte und beginnt mit der Zugabe einer Suspension 

der beschichteten Partikel zu einer verdünnten Probe, woraufhin der nachzuweisende Analyt an die Festphase 

bindet. In einer multiplexen Analysereaktion werden mehrere Partikelpopulationen mit einer verdünnten Probe 

gemischt. Je höher die Konzentration der nachzuweisenden Substanz ist, desto mehr Analyte werden gebunden. 

Im zweiten Schritt wird ein Detektormolekül, in der Regel ein Antikörper mit einer hohen spezifischen Affinität 

zur nachzuweisenden Substanz, zugegeben. Dieses ist wiederum mit einem Fluoreszenzfarbstoff markiert. Die 

Verwendung von Fluoreszenzfarbstoffen mit hohen Quantenausbeuten, wie z. B. Phycoerythrin, garantiert eine 

hohe Empfindlichkeit der Nachweisreaktionen, wodurch die Analyse von Proben mit niedrigem Gehalt oder auch 

von sehr verdünnten Proben möglich wird. In Kombination mit dem multiplexen Testansatz führt dies zu einem 

geringen Bedarf an Probenmaterial.

SERION Multianalyt TM Immunoassay Design 


The analysis is performed in the cavities of a microtiter plate and starts with the addition of a prepared particle 

suspension to a diluted sample preparation. Subsequently, the analyte of interest binds to the solid phase. In a 

multiplex test performance various particle populations are mixed with the diluted sample. The higher the concentration 

of the analyte, the more molecules bind to the capture molecule coated on the surface of the particles. In 

a second step, a detector molecule, usually an antibody with a high specific affinity to the analyte, is added to the 

reaction mixture. The detector is labelled with another fluorescent dye. The use of fluorophores with high quantum 

efficiency, such as Phycoerythrin, guarantees a high sensitivity in the immunoreaction. Thereby, analysis of solutions 

with low analyte concentration as well as highly diluted samples can be performed. The use of a multiplexing 

test system requires very low sample quantities.

Analyse

Partikel einer Population wird das Messergebnis statistisch abgesichert, so dass eine hohe Reproduzierbarkeit 

der erhobenen Daten garantiert wird. Die Fluoreszenzintensität des gebundenen Detektorantikörpers ist 

proportional zur Konzentration des Analyten in der Probe. Da Durchflusszytometer mehrere tausend Partikel pro 

Sekunde analysieren können, erfolgt die Messung einer Probe in nur wenigen Sekunden. 

Analysis 

In a flow cytometer particles are separated and precisely gated through the focus of the laser-based detection 

system. The identification of the particles is performed by the scattering of light and by fluorescence intensity. 

Each particle is analysed individually. The analysis of more than a hundred particles per population assures the 

results statistically and guarantees the high reproducibility of the data. The fluorescence intensity of the bound 

detector molecule is proportional to the concentration of the analyte in the sample. Flow cytometers are able to 

analyse thousands of particles per second. Consequently, the measurement of a sample is performed in a few 

seconds.

Die SERION Multianalyt TM Technologie ist für die Anwendung auf verschiedenen, marktüblichen Durchflusszytometern

Komplementbindungsreaktion 

Die Komplementbindungsreaktion (KBR) ist eine klassische serologische Methode für den Nachweis von 

Antikörpern gegen Erreger von Infektionskrankheiten und hat sich als Standardmethode in der Diagnostik 

vielfach bewährt. Die Institut Virion\Serion GmbH bietet eine breite Palette von Antigenen aus Bakterien, Viren 

und Parasiten für die KBR an, die als Mikromethode nach der KOLMER-Technik entsprechend DIN 58 969 und den 

Empfehlungen der Sachverständigenkommission der WHO durchgeführt wird. 

Complement Fixation Test 

The Complement Fixation Test (CFT) is a classical serological method for the detection of antibodies against 

pathogens of infectious diseases and has proved itself as a standard diagnostic method in many medical 

laboratories. With SERION CFT reagents, Institut Virion\Serion GmbH offers a great variety of antigens that are 

important in infectious serology of bacteria, viruses and parasites. The complement fixation test performance is 

a micromethod according to the KOLMER technique and to DIN 58 969 as well as to the WHO guidelines. 

Testprinzip 

Die SERION KBR beginnt mit der thermischen Inaktivierung von Patientenserumverdünnungen, wodurch Störungen 

des nachfolgenden Reaktionsablaufs durch bereits in den Seren vorhandenes Komplement verhindert werden 

sollen. Durch die Zugabe des KBR Antigens zu den Serumverdünnungen wird die Bildung von Antigen-Antikörper- 

Komplexen eingeleitet. Anschließend erfolgt die Zugabe von Komplement. Die gebildeten Immunkomplexe 

aktivieren das Komplement. Im weiteren Reaktionsverlauf werden sensibilisierte Erythrozyten (Hämolytisches 

System) als Indikator zugegeben. Wurde das Komplement an die Immunkomplexe gebunden, wird der Indikator 

nicht lysiert (Hämolysehemmung). Eine abschließende Zentrifugation der Mikrotiterplatte zeigt bei positiven 

Proben eine Sedimentation der roten Erythrozyten. Bei Abwesenheit von spezifischen Antikörpern im Serum 

bewirkt das Komplement eine Lyse der Schafserythrozyten, wodurch die Bildung eines roten Niederschlags nach 

Zentrifugation ausbleibt und stattdessen ein blass rosa gefärbter Überstand zurückbleibt. 


The first step in the SERION Complement Fixation Tests is the thermal inactivation of patient’s serum samples 

to inactivate endogenous complement which may disturb the test calibration. Antigen is then added, leading to 

the formation of specific antibody-antigen complexes. Subsequent addition of complement will, in the presence 

of antibody-antigen complexes, lead to activation of the complement. An indicator system consisting of antibody 

coated erythrocytes (haemolytic system) is finally added to the system after a suitable time lapse for the 

complement fixation to complete. Lysis of the indicator system is dependent upon the presence or absence 

of immune complexes formed between the sample and specific antigen used. No lysis indicates the presence 

of immune complexes while lysis indicates non. Mild centrifugation of the system sediments any surviving 

erythrocytes, allowing the test to be evaluated visually. A button of red cells indicates a positive result whereas 

a reddish clear solution is negative.

Die KBR kann als Screeningtest für akute Infektionen verwendet werden. Positive KBR Titer sind oftmals ein 

Hinweis auf das Vorhandensein von IgM Antikörpern oder sehr hohen IgG Antikörpertitern und weisen damit auf 

eine akute bzw. erst kurz zurückliegende Infektion hin. Besonders günstig erweist sich die KBR für die Diagnostik 

akuter respiratorischer Infektionen: die in der Regel starke IgG-Boosterung wird in der KBR gut erfaßt, während 

Seronarben vorangegangener Infektionen ausgeblendet werden. Eine Unterscheidung zwischen IgM und IgG 

Antikörpern ist jedoch nicht möglich. 

Diagnostics 

Complement fixation tests can be used as screening tests for acute infections. The fact that one IgM molecule is 

capable of activating one C1-molecule of the complement complex whereas 168 IgG molecules are necessary to 

achieve the same effect, emphasises the significance of the test for the recognition of early infections. Positive 

CFT titres are often an indication for the presence of IgM antibodies or very high IgG antibody titres and are 

therefore an expression for an acute or recent infection, respectively. The CFT is especially helpful for acute 

respiratory infections: The, in general, strong IgG booster reaction is very well represented whereas residual 

titres of past infections are blanked out. The distinction between IgG and IgM antibodies is not possible.

SERION ELISA Analyser 

Der Immunomat TM ist ein Vier-Platten Benchtop-Automat 

mit integriertem Pipettierer, Plattentransporter, ELISA 

Washer und Reader. Vier Stellplätze und Inkubatoren mit 

Schüttelfunktion garantieren eine effiziente Auslastung 

des Geräts, mit dem bis zu sieben Mikrotiterplatten 

bearbeitet werden können. 

The Immunomat TM is a 4-plate benchtop automat with 

integrated pipetting system, plate transporter, ELISA 

washer and reader. Four plate positions and incubators 

with agitation function to guarantee an efficient 

workload of the instrument allowing the processing of 

up to seven microtitre plates.

The Immunomat TM attention to detail includes a barcode identification system for racks, reagents and microtitre 

plates and allows the processing of up to 16 different assays per plate. The easy access to buffers and reagents, 

level sensors for all fluid containers and software-controlled tip management system guarantee customer 

convenient handling. 

Die Immunomat TM Software ermöglicht eine einfache Erstellung von Assayprotokollen, Panel Definitionen und 

Arbeitslisten für Routineanwendungen. Eine Anbindung an Labor-EDV Systeme erfolgt bidirektional über das 

integrierte ASTM Interface. Die Auswertung der Messsignale der SERION ELISA gelingt kundenfreundlich und 

übersichtlich mit Hilfe der Immunomat TM Software. Selbstverständlich entspricht das Gerät den Anforderungen 

der IVD Richtlinie 98/79/EC. 

The Immunomat TM software allows the generation of assay protocols, panel definitions and worklists for routine 

diagnostics. Connection to laboratory software systems is possible via the bi-directional ASTM interface. Evaluation 

of SERION ELISA tests is carried out conveniently by using the Immunomat TM software. Of course, the 

system is in compliance with the in-vitro diagnostic directive 98/79/EC.
