Automated <strong>blood</strong> sampling systems, properly deployed,<br />
can increase the throughput of drug metabolism<br />
and pharmacokinetics research groups. ABS systems<br />
can also improve the accuracy and repeatability of sample<br />
collection, minimize employee exposure to allergens and<br />
reduce the stress on animals caused by handling and restraint.<br />
THREE UNIQUE FEATURES of the INSTECH ABS<br />
1 Undiluted Samples Verified<br />

the <strong>Instech</strong> ABS distinguish<br />
between pure <strong>blood</strong>, diluted<br />
<strong>blood</strong> and IV fluid. Before a<br />
study begins, these sensors<br />
ensure the system is set up<br />

Without analytical guidance, the choice of where to<br />
separate the diluted leading edge from the pure sample<br />
is left to chance. (Visual discrimination is particularly<br />
unreliable.) Trimming too much can lead to wasted<br />
<strong>blood</strong> and overly long sample collection times; too<br />
little, to diluted samples and inaccurate data.<br />

During a study the sensors serve a dual role. First,<br />
they verify that <strong>blood</strong> arrives at the sensors at the<br />
correct time in the sampling process. If not, the<br />
culprit is often a catheter blockage; the system will<br />
respond with an auto-retry routine and alert the user.<br />
Second, the sensors measure the <strong>blood</strong>’s hematocrit,<br />
providing real-time physiologic data.<br />

Other <strong>blood</strong> sampling systems either have no sensors<br />
at all or monitor pressure during the process, a parameter<br />
which is not directly related to sample integrity.<br />
By comparison, the <strong>Instech</strong> ABS monitors the <strong>blood</strong> in<br />
the line to ensure that the <strong>sampler</strong> is set up optimally<br />
before a study, respond to blockages during the study<br />
and provide data for analysis afterward.<br />

2 Direct Path<br />
Peristaltic pumps in the <strong>Instech</strong> ABS move <strong>blood</strong><br />
samples directly from the animal to the vials in<br />
the refrigerated fraction collector. This design has<br />
several advantages over other systems that use a<br />
syringe pump to pull the entire sample back into<br />
reservoir tubing, then push it forward to the<br />
collection vial: (1) the shorter, direct flow path<br />
causes less mixing, which leads to smaller trim<br />
volumes for undiluted samples and in turn to more<br />
samples per animal; (2) sample cycle times are<br />
faster (samples can be taken closer together); and<br />
(3) sample volumes are not limited by the volume<br />
of the reservoir tubing.<br />

3 Flexibility<br />
The <strong>Instech</strong> ABS is designed to work with a wide<br />
range of animals, from mice and rats to large animals.<br />
With mice, the ability to collect small volumes and<br />
minimize waste is critical to avoid excessive hemodilution.<br />
The ABS’s low loss mode returns the <strong>blood</strong> at<br />
the leading edge, cutting wasted <strong>blood</strong> from 40-80µl<br />
to just 3µl per sample. <strong>Instech</strong> Solomon’s tapered 1Fr<br />
FunnelCath and low-torque swivels are also keys to<br />
success with mice.<br />
For large animal sampling, simply switch to larger<br />
catheters and tubing sets. The ABS can even split<br />
a single sample between two collection tubes<br />
when <strong>blood</strong> is plentiful.<br />
Even if you work only with rats, the <strong>Instech</strong> ABS is<br />
remarkably flexible. An <strong>automated</strong> routine calibrates<br />
the system for any catheter you choose. The <strong>sampler</strong>’s<br />
small size and several mounting options give you many<br />
choices for caging, including metabolic cages. Use a<br />
two channel swivel for simultaneous sampling and<br />
infusion or BP measurement, or even a five-channel<br />
swivel for simultaneous sampling and microdialysis.<br />
Automated sampling is an investment. Make the<br />
