Bakteri suşlarının tiplendirilmesi, salgın kaynağının bulunması ve aşı geliştirilmesi
gibi çalışmalara ışık tutmaktadır. Bu çalışmada, tekrar eden DNA dizilerine
(REP-PCR, BOX-PCR) ait primerler kullanılarak Konya bölgesindeki mastitisli
ineklerden izole edilen 98 Staphylococcus aureus suşunun genotiplendirilmesi
amaçlanmıştır. Bakteri suşların tiplendirilmesinde her iki metot önce ayrı
ayrı değerlendirilmiş, daha sonra da iki metodun kombinasyonu alınarak
değerlendirilmiştir. Toplam 44 bant profili Nei homolojisine göre değerlendirilmiş
ve kümeleme analizi (UPGMA) ile bir dendrogram elde edilmiştir. REP-PCR ve
BOX-PCR sonuçlarına göre elde edilen dendrogramlarda, hemen hemen aynı
izolatlar aynı gruplarda yer almışlardır. Bu iki yöntemin kombinasyonu alınarak
elde edilen dendrogramda ise, 98 S. aureus suşunun büyük bir kısmı %62’den
fazla benzerlik göstermiştir. S. aureus suşlarının ayırımını başarılı bir şekilde
sağlayan, kolay, hızlı ve güvenilir olan bu yöntemler, mastitise karşı etkili korunma
yöntemlerinin geliştirilmesinde ve hastalığa neden olan salgının kaynağını bulmak
için tespit edilen patojene göre aşı geliştirilip tedavi edilmesinde kullanılabilir.
Typing of bacterial strains have shed light on studies such as finding source of
epidemics and development of vaccines. In this study, it is aimed to genotype 98
Staphylococcus aureus strains isolated from bovine mastitis in Konya region using
primers belonging to repetitive DNA sequences (REP-PCR, BOX-PCR). At first
both methods in typing bacterial strains were evaluated separately and later the
combination of methods were evaluated. Totally 44 band profiles were evaluated
according to Nei homology and a dendrogram was obtained using cluster analysis
(UPGMA). In the dendrograms, according to the results of REP-PCR and BOX-
PCR obtained, almost the same isolates have been involved in the same groups.
And in the dendrogram based on a combination of these two methods, majority
of 98 S. aureus strains showed more than 62% similarity. These easy, fast and
reliable methods that provide successful distinction of S. aureus strains can be used
to develop effective prevention interventions against mastitis and to develop the
vaccine for the pathogen identified to find the source of the epidemic and to treat it.
