Purging Procedure and a Rabbit Complement<br />

Lysis for Eliminating Burkitt's Lymphoma Cells<br />

Several monoclonal antibodies that react with B lymphocytes are now available<br />

for therapeutic use. We previously showed, according to the immunophenotype<br />

of 28 Burkitt's lymphoma (BL) cell lines, that three of them, and Y29/55—<br />

two pan B reagents—and AL 2<br />

directed against CALLA (common acute lymphoblastoid<br />

leukemia antigen), were expressed on BL cell lines, especially in<br />

Caucasian ones (1 ). We used this cocktail of monoclonal antibodies to target<br />

BL cells, either in a purging procedure using rabbit complement or in an<br />

immunomagnetic depletion technique. A description of how to set up optimal<br />

working conditions in the two techniques has been previously published (2,3;<br />

With an Immunomagnetic Purging Procedure," in this volume). The aim of this<br />

study was, first, to quantify the efficiency of two new IgM monoclonal antibodies,<br />

RFB 7<br />

(CD 20-like) and SB 4<br />

(CD 19), in the rabbit complement lysis procedure<br />

and, second, to compare in the same experiments the efficiency of both<br />

procedures in optimal conditions. Such experiments were performed on three<br />

cell lines freshly established from our patients' tumor cells, all three patients<br />
