SPECIMEN REQUIRED: Refrigerated specimen<br />

COLLECTION INSTRUCTIONS:<br />

1. Pick freshest vesicle<br />

2. Collect culture first, aspirate the fluid from the vesicle<br />

into the tuberculin syringe and inject it into the cell<br />

culture tubes. Obtain collection materials from Virology<br />

M-F 7 am to 5 pm and Saturday 10 am to 12 pm.<br />

3. Unroof vesicle with #15 Bard-Parker scalpel.<br />

4. Scrape the base of the vesicle (obtain basal and parabasal<br />

cells).<br />

5. In the center of 2 glass slides smear the specimen in a<br />

circular motion out to the size of a dime.<br />

6. Air dry and transport to the lab immediately<br />

If Virology is closed use following method for VZ culture only.<br />

1. Unroof vesicle with #15 Bard-Parker scalpel.<br />

2. Scrape the base of the vesicle (obtain basal and parabasal<br />

cells) with a NP/male swab.<br />

3. Break swab in to viral transport media.<br />

4. Transport to the lab immediately. Keep refrigerated.<br />

SPECIMEN REQUIRED: Any specimen source on ice<br />

COLLECTION INSTRUCTIONS:<br />

Nasopharyngeal wash+NP:<br />

1. Draw up 1-2 mL of sterile saline into a 3-5 mL syringe.<br />

2. Remove the needle from the syringe and put on a butterfly<br />

with a large tubing. Cut the tubing to a length of 2-3".<br />

3. Place the tubing in the nasopharyngeal passage. Inject the<br />

saline rapidly and draw back rapidly.<br />

4. Inject saline and nasal material into the viral transport<br />

medium. If viral transport medium is not available, the NP<br />

wash may be added to 1.O mL of trypticase soy broth or<br />

sterile saline.<br />

5. After or before doing the wash, use a dacron NP swab to do<br />

a good epithelial cell scraping of the nasopharyngeal area<br />

Place and cut or break the swab into the same vial as the<br />

wash.<br />

TUBE OR CONTAINER: Viral transport tube, Lavender (EDTA) or<br />

Sterile container (urine)
