■ anticipating expected and unintended adverse effects <str<strong>on</strong>g>of</str<strong>on</strong>g> all experimental work;<br />

■ being familiar with subtle signs <str<strong>on</strong>g>of</str<strong>on</strong>g> distress or discomfort in the species, strain, phenotype<br />

and individual animal, and knowing how to alleviate the cause;<br />

■ disseminating specific informati<strong>on</strong> <strong>on</strong> Refinement in an accessible way;<br />

■ publishing details <str<strong>on</strong>g>of</str<strong>on</strong>g> Refinement as an integral part <str<strong>on</strong>g>of</str<strong>on</strong>g> scientific papers in the<br />

mainstream literature; and<br />

■ most importantly, not assuming that existing practice is necessarily best practice.<br />

Summary<br />

12.29 Effective implementati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> Refinement and Reducti<strong>on</strong> requires both c<strong>on</strong>cepts to be<br />

c<strong>on</strong>sidered at an early stage to improve the general <str<strong>on</strong>g>research</str<strong>on</strong>g> strategy <str<strong>on</strong>g>of</str<strong>on</strong>g> a project. A staged<br />

approach before embarking <strong>on</strong> large or complex experiments is useful, starting with<br />

thorough background <str<strong>on</strong>g>research</str<strong>on</strong>g> <str<strong>on</strong>g>of</str<strong>on</strong>g> the published literature and c<strong>on</strong>sidering the possibility <str<strong>on</strong>g>of</str<strong>on</strong>g><br />

c<strong>on</strong>ducting a small pilot study. Effective teamwork also plays a significant role, <str<strong>on</strong>g>involving</str<strong>on</strong>g><br />

staff with a wide range <str<strong>on</strong>g>of</str<strong>on</strong>g> relevant expertise, in for example in vitro technology,<br />

experimental design, statistics, and animal care.<br />

12.30 We observed that the c<strong>on</strong>cept <str<strong>on</strong>g>of</str<strong>on</strong>g> Reducti<strong>on</strong> is best understood as requiring ‘the use <str<strong>on</strong>g>of</str<strong>on</strong>g> fewer<br />

<str<strong>on</strong>g>animals</str<strong>on</strong>g> in each experiment without compromising scientific output and the quality <str<strong>on</strong>g>of</str<strong>on</strong>g><br />

biomedical <str<strong>on</strong>g>research</str<strong>on</strong>g> and testing, and without compromising animal welfare’. To improve its<br />

applicati<strong>on</strong>, the importance <str<strong>on</strong>g>of</str<strong>on</strong>g> appropriate <str<strong>on</strong>g>research</str<strong>on</strong>g> strategies, better c<strong>on</strong>trol <str<strong>on</strong>g>of</str<strong>on</strong>g> variati<strong>on</strong><br />

am<strong>on</strong>g <str<strong>on</strong>g>animals</str<strong>on</strong>g>, better statistical analysis and the avoidance <str<strong>on</strong>g>of</str<strong>on</strong>g> duplicati<strong>on</strong> need to be<br />

recognised. We c<strong>on</strong>sidered successful examples <str<strong>on</strong>g>of</str<strong>on</strong>g> Reducti<strong>on</strong> in regulatory testing and<br />

noted that harm<strong>on</strong>isati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> internati<strong>on</strong>al test guidelines can c<strong>on</strong>tribute significantly to<br />

further reducti<strong>on</strong>.<br />

12.31 Refinement is probably the most effective <str<strong>on</strong>g>of</str<strong>on</strong>g> the Three Rs in achieving immediate reducti<strong>on</strong><br />

<str<strong>on</strong>g>of</str<strong>on</strong>g> pain and suffering, and improvement <str<strong>on</strong>g>of</str<strong>on</strong>g> welfare <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>animals</str<strong>on</strong>g> involved in <str<strong>on</strong>g>research</str<strong>on</strong>g>. <str<strong>on</strong>g>The</str<strong>on</strong>g><br />

approach is <str<strong>on</strong>g>of</str<strong>on</strong>g> great relevance since reducing pain, suffering and distress is a crucial aspect<br />

<str<strong>on</strong>g>of</str<strong>on</strong>g> the moral debate about animal <str<strong>on</strong>g>research</str<strong>on</strong>g>, and a legal requirement. It is also important<br />

scientifically since the physical and psychological welfare <str<strong>on</strong>g>of</str<strong>on</strong>g> laboratory <str<strong>on</strong>g>animals</str<strong>on</strong>g> can have a<br />

significant effect <strong>on</strong> the scientific validity <str<strong>on</strong>g>of</str<strong>on</strong>g> experimental results.<br />

12.32 Possibilities for implementing Refinement were c<strong>on</strong>sidered in four areas: housing,<br />

husbandry and care, experimental procedures, pain management and humane endpoints.<br />

Refinement <str<strong>on</strong>g>of</str<strong>on</strong>g> housing c<strong>on</strong>diti<strong>on</strong>s is particularly important since the quality <str<strong>on</strong>g>of</str<strong>on</strong>g> <str<strong>on</strong>g>animals</str<strong>on</strong>g>’ cage<br />

or pen envir<strong>on</strong>ments can have a major impact <strong>on</strong> their lives. While standards for laboratory<br />

housing are defined in the Home Office Codes <str<strong>on</strong>g>of</str<strong>on</strong>g> Practice for the husbandry and care <str<strong>on</strong>g>of</str<strong>on</strong>g><br />

<str<strong>on</strong>g>animals</str<strong>on</strong>g> and relevant European guidelines, the requirements represent minimum standards.<br />

<str<strong>on</strong>g>The</str<strong>on</strong>g>re should be relatively few scientific barriers to Refinement, and these should be<br />

c<strong>on</strong>sidered <strong>on</strong> a case by case basis. A fundamental principle is never to assume that current<br />

practice is best practice. All the techniques and protocols that are used at regular intervals<br />

should be reviewed and critically assessed throughout the durati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g> any <str<strong>on</strong>g>research</str<strong>on</strong>g><br />

programme. We present our c<strong>on</strong>clusi<strong>on</strong>s and recommendati<strong>on</strong>s about the implementati<strong>on</strong><br />

<str<strong>on</strong>g>of</str<strong>on</strong>g> the Three Rs in Chapter 15 (paragraphs 15.57–15.62) and now turn to the regulati<strong>on</strong> <str<strong>on</strong>g>of</str<strong>on</strong>g><br />

animal <str<strong>on</strong>g>research</str<strong>on</strong>g>.<br />
