Section 2 Preparation<br />
10.After equilibration, the Torrent will pause for sample<br />
injection. Draw 2 mL of the dissolved Test Mix B solution<br />
into a syringe and inject it:<br />
a. Place the syringe on top of the Liquid Injection<br />
Accessory valve.<br />
b. Open the valve by turning the handle so it is in line with<br />
the tubing.<br />
c. Inject the liquid.<br />
d. Close the valve by turning the handle so that it is<br />
perpendicular to the tubing. Remove the syringe.<br />
Note<br />
A chase solvent forces the sample through the automated<br />
injection valve.<br />
11.Inject 10 mL of ethyl acetate as a chase solvent:<br />
a. Draw 5 mL of ethyl acetate into the syringe and inject<br />
this as described in the sub-steps of step 10 above.<br />
b. Repeat step a for a second 5 mL injection (10 mL total).<br />
12.Touch the OK button to continue the run.<br />
13.While the Torrent is operating, observe the Fraction<br />
Collector/Fractionation Valve operation. Peaks should be<br />
cut and collected in tubes/containers. Waste fluids should<br />
be diverted to the waste container.<br />
14.When the Torrent finishes the run, review the resulting<br />
chromatogram which should meet the following criteria<br />
(refer to Figure 2-22):<br />
• Baseline must not rise more than 0.1 AU during<br />
separation.<br />
• Height of Peak 2 must be higher than Peak 3.<br />
• Retention time of Peak 2 (time that the top of the<br />
second peak occurs) must be between 5.5 and 7.5<br />
column volumes.<br />
If the criteria is not met, repeat the verification steps. If the<br />
results still do not meet the listed criteria, contact your<br />
Teledyne <strong>Isco</strong> representative.<br />
