4.4.2.1 Basic method settings<br />

The basic settings allow you to control the %B gradient mix,<br />

flow rate, solvents, and run length. These settings are part of<br />

the MAIN WINDOW [4.3.1]. These settings are the most<br />

frequently changed settings when developing or improving<br />

purification methods.<br />

The %B gradient mix and run length are shown on the plot<br />

area. Refer to the DEFINING A GRADIENT [4.4.5] topic for more<br />

details. The other settings are:<br />

• Flow Rate — When using the <strong>Isco</strong> default column<br />

methods, the flow rate is automatically set to the<br />

optimum flow rate of the column. If using a non-<strong>Isco</strong><br />

column, refer to the manufacturer’s column<br />

documentation to determine the best flow rate for the<br />

column. Note that if the column is not detected by the<br />

module, the operating pressure is limited to 50 psi.<br />

• Solvent A and B — Select the desired solvents from the<br />

lists. The solvents are those defined by the<br />

CONFIGURATION [4.3.10] window.<br />

4.4.2.2 Advanced method settings<br />

The METHOD EDITOR [4.3.2] window gives you access to<br />

advanced settings such as equilibration volume, initial waste,<br />

air purge, peak detection, and peak collection mode.<br />

Equilibration Volume Column equilibration is optional, but<br />

you may find that separations are more effective if the column<br />

is equilibrated before injecting the sample. The recommended<br />

equilibration volume is at least three column volumes of the<br />

starting mobile phase.<br />

RediSep Rf Silica gel column volumes are:<br />

• RediSep Rf 80 g = 125 mL<br />

• RediSep Rf 120 g = 192 mL<br />

• RediSep Rf 220 g = 334 mL<br />

• RediSep Rf 330 g = 443 mL<br />

• RediSep Rf 750 g = 1.2 L<br />

• RediSep Rf 1.5 kg = 2.4 L<br />
